Abstract:Acetyl xylan esterase(Axe) can hydrolyze O-acetyl located in acetylated xylan. Thus,the steric hindrance effects of O-acetyl can be eliminated when xylan is hydroyzed by xylanase. In this study,a mature peptide gene,Auaxe,encoding an acetyl xylan esterase(AuAXE),was amplified by RT-PCR technique using the total RNA from Aspergillus usamii E001 as template. The recombinant expressing plasmid pPIC9KM-Auaxe was constructed and linearized with SalⅠ,followed by transforming it into Pichia pastoris GS115 by electroporation. The recombinant P. pastoris was screened by G418,induced by methanol for 72 h,and then the recombinant acetyl xylan esterase(reAuAxe) was obtained. The activity of crude reAuAxe reached 35.6 U/mL. After purification,the reAuAxe was purified to electrophoretic homogeneity with the activity of 390.5 U/mg. SDS-PAGE analysis showed that the apparent molecular mass of purified reAuAxe was about 34.0 kDa. Additionally,the enzymatic properties of purified reAuAxe were also analyzed. Its optimal temperature and pH were 50 ℃ and 6.0,respectively. It was stable at a temperature of 45 ℃ or below,and at a pH range of 4.5~7.0. Its enzymatic acvitity was not signficantly affected by an array of tested metal ions and EDTA. This paper laid a solid theoretical foundation for this enzyme's study in depth and application.

Abstract:Based on transcriptome Unigene sequence the diacylglycerol Kinase gene named CoDGK3(GeneBank accession number:KM068056) was isolated from Camellia oleifera with the method of RT-PCR and RACE. Bioinformatics approaches are used to analyze and predict the structural characters and physicochemical property. The bioinformatics analysis showed that the CDS of CoDGK3 is 1568 bp in length,OFR 1455 bp and it codes predicted protein of 485 amino acids. Homologous comparison and sequence analysis of CoDGK3 amino acids suggested that the CoDGK3 is highly conserved and showed the highest similarity(78%) with the DGK of Jatropha curcas and Populus trichocarpa.The gene expression changes by qPCR were similar to those of RTKM in transcriptome data indicate that the CoDGK3 expresses stably in different developmental stage of Camellia oleifera seeds and the transcriptome data of Camellia oleifera seeds is valid and reliable.

Abstract:For the purpose of improving the production of glucose oxidase,response surface methodology(RSM) has been employed to optimize the fermentation medium to improve glucose oxidase production by mutational Aspergillus niger. Firstly,a Plackett-Burman design was used to investigate the effects of different factors in the fermentation medium,with three most significant factors being identified as:beef extract peptone,Tw-60 and(NH4)2HPO4. Secondly,the steepest ascent procedure was employed to define the optimal response region for these three factors. Finally,the Box-Behnken method was employed to design the reaction system and the optimal fermentation medium for production of glucose oxidase consisted of the following(g/L):sugar 87.5,beef extract peptone 3.15,NH4NO3 1.88,(NH4)2HPO4 0.34, KH2PO4 0.25,Tw-60 30.47,corn flour 12.5. Glucose oxidase production after optimization was increasing by a factor of about 93.28% from 45.27 U/mL to 87.5 U/mL. The different adding time of Tw-60 was investigated in 7 L fermentor. The result shows that adding Tw-60 to broth in logarithmic phase can improve glucose oxidase production remarkably,the total glucose oxidase activity reached 92.88 U/mL.

Abstract:The effects of drying and extraction processes on the antioxidant compounds and antioxidant activities of one kind of yam(Dioscorea alata), were investigated. The antioxidant compounds analyzed the phenolic content. The antioxidant measurements included a ,a-diphenyl-b-pricryl-hydrazyl(DPPH) radical-scavenging activity and reducing power . The 50% ethanolic extracts from the freeze-dried peel had the most antioxidant compouds and the highest antioxidant activities. Generally speaking,freeze-dried yam maintained higher antioxidant activities than hot air-dried ,the peel had more than the meat,Among three extraction methods,50% ethanolic extraction was the best.By HPLC analysis showed that the major polyphenols of the extraction was chlorogenic acid. Meanwhile tested the 50% ethanolic extracts on the inhibition of the formation of ROS,we got the same resulet as before.

Abstract:This paper studied the preparation technology of fat substitute on the basis of indica rice. In this paper,with the method of single factor experiment,the influence of dextrose equivalent of the products(DE) on the alpha-amylase additive volume resistant to the high temperatures,indica rice flour mesh and hydrolysis time was researched. Furthermore,the optimum technological condition of indica rice based fat substitute was determined by orthogonal test,conditions were as follows:enzyme additive volume was 4.0 U/g rice flour,hydrolysis time was 30 min,indica rice flour mesh was 100 mesh. The DE value of the product prepared was around 3.0,and the dried product was white powder. The precipitation of extracted fat substitute was utilized and continued to prepare the maltodextrin with DE value of 13~17. These technological conditions were enzyme additive volume was 40.0 U/g rice flour,hydrolysis time was 40 min. Finally,the results showed that the fat substitute of 6% quality fraction and 9% maltodextrin were compounded to give the instant coffee with the best taste.

Abstract:The alterations of papain activity and conformation were studied in four kinds of organic solvent,which are propanediol,glycerol,propanol and ethanediol. The result showed that the papain activity increased 20.1% in 15% propanediol,increased 17.8% in 8% propanol and decreased obviously in glycerol and ethanediol. The value of Km and Vmax was declined when treated with four kinds of organic solvent. Ultraviolet difference spectra and Fluorescence emission spectra showed that the secondary structure of papain altered in four kinds of organic solvent. The papain structure containning Trp residue was in a stronger non-polar environment in 40% glycerol,while the papain structure where coil and sheet increase contains little Trp residue in 15% propanediol and 10% ethanediol.

Abstract:Utilize single pollution index and international health risk assessment method recommended by the health status of a commercial part of the risk of contamination of leafy vegetables to their lead and cadmium in the sample cause analysis and evaluated. leafy vegetables cadmium content of lead exceeding the rate of 12.39%;single pollution index values were lower than 2.0;Pb is 3.99×10-8(a-1)~1.80×10-7(a-1),the sum of 9.55×10-7(a-1).The annual average value range of personal health risks Cd is 1.53×10-5(a-1)~4.09×10-5(a-1),the sum of 2.13×10-4(a-1). The commercial part leafy vegetables Pb exceeded;single pollution index shows that the results of the evaluation of commercially available leafy vegetables are excellent and basic safety range;from health risks index,the commercially available lead and cadmium in leafy vegetables hazards to human health personal annual risk at a safe level.But it was a healthy sum of the individual risk than ICRP recommended standard values.This shows that the city's residents dietary intake of vegetables,Pb,Cd has a potential health threat.
