Abstract:Endothelial apoptosis which is an early factor causing endothelial injury,plays an significant role in the early stage of cardiovascular.At present,the effects of TFA on atherosclerosis and othe cardiovascular diseases at home and abroard mainly focus on endothelial cell apoptosis.The were little research of the realationship between TFA and human health in spite of the TFA content of chinese food rising year after year.At present in the fields of domestic research is still in its beginning stage.There were little conculted literature review limitting to the relationship of TFA and huamn health.The mechanism of TFA on endothelial cells mainly focus on endothelial apoptosis signal transduction.The study of the effects of TFA on the endothelial cells was of an important theoretical and social value,and provides theoretical basis and reliable scientific data for the prevention and treatment of cardiovascular diseases.

Abstract:Effect of 1-octylcclopropene(1-OCP) concentrations at room temperature for 20 h on the storage quality of post-harvest ‘Guifei’ mangoes(Mangifera indica L.) was investigated with comparison of the treatment with same concentrations of 1-MCP,a reported effective ethylene action inhibitor.Fruits were fumigated with 1 μL/L,5 μL/L or 50 μL/L 1-OCP or 1-MCP and then stored at low temperature(4±1 ℃,RH85%-90%) with every 3 fruits packed in a 0.02mm thick PE bag(240 mm ×190 mm) for ripening evaluation.Quality changes and relevant physiological responses were measured every 3 days during fruit storage.The results show that treatments with 50 μL/L 1-OCP or 5 μL/L 1-MCP significantly delayed the appearance of respiratory rate peak,fruit softening and decrease in titratable acid,postponed fruit weight-loss and increase in soluble solids content,inhibited PPO(polyphenol oxidase) and POD(peroxidase) activity and MDA accumulation.

Abstract:Effect of the aqueous fraction from ethanolic extract of Boschniakia rossica(BRAF) on the oxidative stress and apoptosis of liver in mice with an acute liver injury induced by Lipopolysaccharide(LPS) and D-galactosamine(GalN) was investigated in this manuscript.The mice were randomly assigned to 5 groups: normal control,model control,silymarin(positive control),as well as high and low dose groups of BRAF.Animals were treated once daily for 7 days.At the end of the experiment,LPS and GalN were given intraperitoneally to the mice of groups,then the pathological changes was detected with hematoxylin & eosin(HE) staining,tumor necrosis factor-α(TNF-α) was detected by ELISA method,the alanine aminotransferase(ALT),aspartate aminotransferase(AST),catalase(CAT),glutathione peroxidase(GPx),glutathione S-transferase(GST),reduced glutathione(GSH),malondialdehyde(MDA) and nitric oxide(NO) were assayed with colorimetric method,and DNA damage was analyzed using the electrophoretic method.The results showed that the administration with BRAF reduced the serum ALT and AST,alleviated the hepatic lesions induced by GalN/LPS,decreased serum TNF-α level,reduced the hepatic NO and MDA contents,increased the hepatic CAT,GPx,GST and GSH levels,and reduced the DNA damage of hepatocyte in mice with acute liver injury.It is suggested that BRAF has a protective effect on acute liver injury induced by GalN and LPS in mice,probably via antioxidative and anti-apoptotic abilities.

Abstract:Effect of twice treatment with three cyclopropene ethylene inhibitors with different 1 substituted branch chain lengths on postharvest ethylene biosynthesis and gene expression of tomato fruits(Solanum lycopersicum L.) were investigated in this manuscript.The fruits were packed with plastic bags(0.02 mm) after being fumigated for 20h with 0.75 μL/L 1-MCP(1 Methylcyclopropene),2 μL/L 1-PentCP(1-pentylcyclopropene) or 0.5 μL/L 1-OCP(1 octylcyclopropene) and then stored at ambient temperature(20 ℃) for maturation evalution,and second time treated wound be in 8 days as the first.The results show that treatments with any one of these three inhibitors,compared with that of the untreated fruits,reduced ethylene production and respiration rate were decreased and,the express level of ACS2 and ACO1 gene were lower in the fruits.Furthermore,these effects tended to decrease as the 1-substituted branch chain length of the cyclopropenes increased.In addition,treatments with any inhibitors delayed physiological metabolism and maintained fruit quality.Twice treatments,however,did not exert obvious effects.

Abstract:Microbial diversity of mixed culture can result in substantial heterogeneity in physiological and biochemical behaviors such as responses against stress.Such differences can cause "cask effect" during H2 production,and thereby restrict the overall production capacity.In this study,butyric acid stress combined with selective chemical inhibitors was used as the selection pressure to evolve mixed culture in continuous operation.By the adaptive evolution,H2 production capacity via mixed culture can be enhanced by removing the bacteria causing "cask effect" and enriching functional H2-producing bacteria obtaining beneficial evolution.The enhancement was primarily driven by increased acid tolerance,weakened competing pathways,higher activity of hydrogenase and NADH-ferredoxin oxidoreductase(NFOR).

Abstract:To study the possible mechanisms of Pomegranate Peel Polyphenols、Punicalagine and ellagic acid on cholesterol biosynthesis.The model of steatotic L-02 hepatocyte was established by cultured with 50%(V/V) fetal bovine serum for 48 hours,and the appropriate concentrations of PPPs were detected by MTT assay,Then the experiment is divided into there groups:normal group、 model group and therapy group,different contents of PPPs is added to the therapy group.After 48h cultured,the lipid droplets stained with oil red O were observed,meanwhile,the intracellular total cholesterol levels were determined by HPLC and the activity of HMG-CoA reductase was detected by ultraviolet spectro-velocity method.The accumulation of lipid droplets and the content of TC were decreased in PPPs treatment group,existing dose-effect relationship,meanwhile,the activity of HMG-CoA reductase was significantly inhibited in PPPs treatment groups,especially in the Punicalagin group.PPPs can decrease the content of TC in steatotic hepatocytes,The possible mechanism is by downregulating the activity of HMG-CoA reductase,Punicalagin was the key component in PPPs on lipid-lowering.

Abstract:The aim of this manuscript was to investrated the effect of some factors,including the ozone treatment time,the sample moisture content and the unit volume sample quality on the the degradation AFB1 by single factor experiment and response surface methodology analysis.The optimum process conditions were achieved and listed as follows: ozone treatment time 7.92 min,sample moisture content 15%,the quality of the sample 5 g.With the optimum conditions,the the ozone detoxification rate can reached at 85%.

Abstract:This work set up a new naked eye detection method for FB1 abased on the high identification function of DNA aptamer and the color changing effect of AuNPs.This assay method taked AuNPs as carrier and firstly linked to DNA1 and then assembled FB1-aptamer-DNA1-AuNPs complex throuth the hybridzation process between FB1-aptamer and DNA1-AuNPs.When added different concentrations of FB1,FB1-aptamer prefered to connect to FB1 and then subsequently additional DNA2 chain that matched with DNA 1 chain can hybrid with exposed DNA1 resulting in AuNPs aggregation.Thus the color changes from red to purple and blue could be observed by the naked eye.Optimal conditions avoided to much salt induced AuNPs aggregation caused error effectively.Furthermore,we added SDS in DNA1 or DNA2 chain connected to AuNPs to maintain the color of AuNPs and improved the concentration of NaCl to 500 mmol/L and DNA loading enlarge 3 times.This breaks the traditional boundary of 100 mmol/L NaCl can caused the color change of AuNPs.The linear range of the colorimetric aptasensor covers a large variation of FB 1 concentration from 125 ng/L to 1500 ng/L and the detection limit of 125 ng/L is obtained.

Abstract:The aim of this study was to develop a rapid quantitative melamine ELISA test kit for the detection of Nisin in dairy,meat and milk drink.The indirect competitive enzyme-linked immunosorbent assay was established based on the Nisin specific polyclonal antibody.The sensitivity was 0.1 μg/mL,the standard curve range was 0.1~62.5 μg/mL,the detection limit of the dairy and meat were 1.0 μg/mL,and 0.5 μg/g,respectively.The average recoveries of milk drink and meat ranges between 80% ~120%,and the coefficient of variation was <20%.The indirect competitive ELISA has been proven to be rapid,sensitive,and simple.It is suitable for the detection of the Nisin content in dairy,meat and milk drink.

Abstract:α-Acetolactate decarboxylase(ALDC) gene alsD was cloned from Bacillus subtilis L520 and constructed into pMA5.The recombinant plasmid pMA5-alsD-His was transformed into B.subtilis WB600.SDS-PAGE indicated that alsD gene was successfully expressed in recombinant B.subtilis and the molecular mass of ALDC was 32×103.Recombinant protein was purified by Ni-NTA affinity chromatography.The specific activity and total yield of the purified enzyme were 272.3 U/ mg and 89.8%,respectively.The optimum temperature of purified ALDC was 25 ℃.And the enzyme showed high activity between 20 ℃ and 35 ℃.The enzyme activity was activated by Ni2+ and totally inhibited by Fe3+.When cultured in shake-flask,the ALDC activity of the recombinant strain was approximately 70-fold higher than that of original strain.The result of scaled-up experiment showed that the enzyme activity was 75.9 U/mL in 5 L fermentor.

Abstract:In this manuscript,the hapten for paraquat N-(4-carboxybutyl)-N’-methylbipyridilium(named PQ-h) was synthesized by using 4,4’-Bipyridine and iodomethane as substrate through three steps.The products was further identified through LC/MS,1HNMR and IR.This study lays the groundwork for further research of immunoassay to paraquat.

Abstract:The aims of this manuscript were to study arsenic speciation and content in kelp with different processing methods(direct drying,blanching,blanching-salting-washing) by high performance liquid chromatography coupled to inductively coupled plasma-mass spectrometry(HPLC-ICP-MS).The results showed that the total arsenic content in direct drying kelp was 47.23 mg/kg and blanching kelp was 42.91 mg/kg(dry weight) with no significant differences(P>0.05).The total arsenic content in blanching-salting-washing kelp was 25.62 mg/kg(dry weight),there was extremely significant differences(P<0.01) from direct drying kelp and blanching kelp.The total arsenic content in kelps was gradually decreased after blanching-salting-washing,while the saltingwashing process decreased most obviously.Arsenocholine,arsenobetaine,dimethylarsinic acid and three substances suspected as arsenicsugar were the major arsenic forms in kelps,which arsenicsugar was in the hightest amount(w>95%) and inorganic arsenic was in the lowest amount(w<1%).

Abstract:In this study,a genomic library for Pichia stipitis was firstly constructed.The library covered 1.08 times of the15 441 179bp genomic DNA of the strain,and contained 3 000 clones with the insertion sizes ranging from 0.5 to 8.0 Kb.It has a propability of 97% for screening a random gene form this library.The library plasmids were extracted and transformed into Saccharomyces cerevisiae W303-1A by LiAc method.At last,a 1.82 Kb cellobiase gene of P.stipitis was firstly characterized by means of screening the transformants on cellobiose plates.

Abstract:To observe the effects of different fermentation days of Shuanqian Junzhi(SQJZ,a Strychnos nux-vomica product fermented by Trametes cinnabarina) on acute toxicity and analgesic,anti-inflammatory activity,seven samples of different fermentation days(0 d,7 d,20 d,41 d,48 d,62 d and 69 d) were applied.The acute toxicity and anti-inflammatory,analgesic activities of SQJZ of different fermentation days were evaluated in animal models.Listed as followed was the acute toxicity of different SQJZ in a descending order:0 d≈7 d <20 d≈41 d <48 d <62 d <69 d.The sustaining twisting reaction times was:69 d≈62 d>48 d>41 d≈20 d>0 d>7 d.According to the control group,the twisting times caused by acetic acid of the crude,0 d,7 d,20 d,41 d,48 d of SQJZ noticeably decreased(P<0.05) just like the crude Strychnos nux-vomica,which indicated the analgesic action of the crude did not change significantly within 48 day-fermentation period.However,62 d and 69 d of SQJZ had no significant analgesic effect and there was a significant difference(P<0.01) in comparison with the positive drug indometacin,which showed a complete loss of the anti-inflammatory effect after 62 day-fermentation.The swelling percent decrease of external ear was:7 d≈20 d>41 d>48 d>0 d>>62 d>69 d.With the control group,0 d,7 d,20 d,41 d and 48 d of SQJZ were able to reduce the swelling of murine external ear caused by croton oil(P<0.05 or 0.01),the same as the crude,indicating that their anti-inflammatory effect did not declined through 48 day-fermentation.Therefore,the feasible period of fermentation of SQJZ is about 41-48 days.

Abstract:The aim of this manuscript was to investigate the effects of taurine on iron utilization and antioxidative ability of iron deficiency anemia(IDA) rats and normal rats,for those,the iron utilization and antioxidative ability in IDA rats and normal rats were determined after gavaged 0.5 g/kg taurine for 21 days.The IDA rat model was established through fed iron deficiency feed to the SD rats Hemoglobin,serum iron,serum ferritin,total iron binding capacity and the content of liver and spleen iron,MDA,SOD,GSH-Px and T-AOC were determined respectively after the end of the intervention.It was found that the addition of taurine in IDA rats increased hemoglobin,serum iron,serum ferritin,the content of liver iron and decreased the total iron binding capacity significantly(P < 0.05).Adding taurine improved the antioxidative ability in IDA rats,especially when adding iron in the same time.Adding taurine had no significant effects on iron utilization and antioxidative ability in normal rats(P > 0.05).Therefore,the effects of taurine on iron utilization and antioxidative ability in the IDA rats were better than that of the normal rats.

Abstract:To study the antibacterial activity of grape seed extract(GSE) constituents(most of them are polyphenolic compounds) against periodontitis and cariogenic bacterial,the relationship between the structures and the antibacterial activities was investigated with optical density(OD) test and the minimum inhibition concentration(MIC) as index.As the result,the fraction with high polarity which is very dissoluble in water has stronger inhibition ability than other fractions with lower polarity.Among the water dissoluble fraction,those compounds with three consecutive hydroxyl groups seems to be very important to antibacterial activity.That is to say,the inhibition factors in the GSE are those phenolic compounds with three consecutive hydroxyl groups but not the procyanidins as what we expect.

Abstract:The objective of this study is to study relieving a cough and anti-inflammation effects of different extract part from Reineckia carnea.For this,the different extract part of Reineckia carnea were orally administrated to coughing mice induced by ammonia liquor in mice and xylene-induced mouse ear swelling model in mice,respectively,to investigate their medication effect on coughing and anti-inflammation.It was found that both ethanol extract and water extract had obvious effect on anti-inflammation(P<0.05),ethanol extract lengthened the latent period of cough at high and low dose(P<0.05),and high dose decrease cough frequency(P<0.05).The water extract fraction lengthened the latent period of cough as well as the ethanol extract at high dose.The petroleum ether extract had not obvious effect on anti-inflammation and coughing.Based on the aboved results,it can concluded that the anti-inflammatory,cough active chemical composition were mainly concentrated in the consumption of alcohol extract parts and water extract high doses parts.
