Abstract:In this manuscript,the progress and deficiency of drug metabolism of traditional Chinese herbs were introduced.The technical advantages were pointed out that drug metabolism research was performed by using the microbial models which provide a novel ways for Chinese herbs drug metabolism.Based on the analysis of practical meaning and prospect of the special ’bi-directional fermentation’ technology,the assumption of establishing the microbial models of drug metabolism of Chinese herbs was suggested by applying this technology.

Abstract:Pycnoporus sanguineus SYBC-L12 has strong ability to produce high level of laccase using rice straw as substrate and support materials under solid-state fermentation.We optimized the condition for laccase production using one-factor-at-a-time method and the laccase activity of the optimized condition was about 10 times higher than that of the original level.The obtained laccase was used to decolorize anthraquinone dyes,triphenylmethane dyes and ethyleniminoquinone dyes and their mixed one at room temperature,which showed desirable results.Under higher temperature,azo dyes were partly decolorized.Moreover,most mental ions of industrial effluent used in the essay had no inhibitory effect on the decolorization except a few.Therefore,laccase as a kind of environmentally friendly bio-catalyzer has a great potential in sewage disposal and effluent treatment.

Abstract:An effective two-dimensional electrophoresis system was developed to determine the total protein from "huiyulu" peach fruit in this study.The optimum conditions for total protein detection were listed as follows: phenol extraction,800 μg protein sample,17 cm IPG strip with pH 4-7,and finally detecting proteins with modified colloidal coomassie brilliant blue G-250 staining.With the optimum conditions,a high quality 2-DE map with low background was achieved.

Abstract:Combining Quantum dot fluorescence labeled technique,magnetic separation technology and competitive immunoassay technology,a new immune detection method of aflatoxin B1(AFB1) was constructed.Primarily,the cadmium telluride quantum dots(CdTe QDs) were prepared in aqueous phase,magnetic nanoparticles were prepared by using hydrothermal synthesis,and characterized by TEM,fluorescence spectra,XRD and Infrared spectroscopy.Then taking AFB1 artificial antigen functionalized magnetic nanoparticles as capture probes,Luminescent quantum dot labeled antibody as signal probes,based on the competitive immune principle,established a novel detection method for AFB1.Under the optimum conditions,the fluorescence intensity was linear in the range of 0.1 ng/mL~100 ng/mL of standardized AFB1 with the detection limit of 0.03 ng/mL.This method has been proven to be higher accuracy.

Abstract:The aim of this study is to determine the building conditions for developing immunomagnetic bead using human immunoglobulin G enrichment of Staphylococcus aureus.For this,the preparation conditions of immunomagnetic beads was optimized by matrix method and the single factor,and the the degree of fixed antibody to immunomagnetic bead was evulated by the amount of saturated adsorption capacity and correlation.Inspect enrichment ability of immunomagnetic bead to Staphylococcus aureus according to specificity,sensitivity and saturation catches.Aas a result,the homemade IMBS.aureus was obtained.The bead was adopted the type of MB-M,added activator combination EDC and NHS(5 mg/L),then incubated 24 h with human immunoglobulin G at 24 ℃;Adsorption capacity of IMBS.aureus was 393.27 g;There was correlation between the contents of beads and human immunoglobulin G;IMBS.aureus has high specificity,sensitivity(26 CFU/10 mL) and catch capability.The aboved results demonstrated that Homemade IMBS.aureus exhibited the characters of stable property,strong accumulation ability and low costs.This manuscript provided a novel way to deal with rapid detection about food-borne pathogens.

Abstract:UHT milk can be clotted or hydrolyzed by the heat-stable protease from Psychrotrophs bacteria in raw milk,resulting in bitter taste and other quality deterioration in shelf life.One of bacterium from raw milk was identified based on morphology,physiology,biochemistry analysis,16S rDNA and 16-23S rRNA sequence analysis.The bacterium(named Rm12) was identified as Pseudomonads fluorescens.The optimal catalytic temperature and pH of the protease from Rm12 were 40 ℃ and 6.8 respectively.After heat treatment(140 ℃,4 s),the rest of activity of protease was only 6.66% that of the original preparation.

Abstract:Regression analysis and statistical methods were used to establish the optimal model of the chemical content and smoke composition.The results show that:1) equations were examined in full model,double screening stepwise regression,and stepwise regression only containing relevant variable.However,the prediction accuracy of the model was different in these methods,and not all the regression coefficient of the predictor variables was examined.2) when the indistinctive variable removed,the higher prediction accuracy and regression diagnostics models about Tar,nicotinamide and CO were obtainted.Tar regression equation was Ytar=11.7+1.857Xnicotine,Nicotine regression equation was Ynicotion=0.679Xnicotion,CO regression equation was YCO=1.428 6Xnicotion+0.121 7Xreducing sugar+2.822 5Xtotal chlorine+8.488 9,and all the regression equation test were significant(P<0.01).

Abstract:The-cyclodextrin glycosyltransferase(α-CGTase) from Paenibacillus macerans,Escherichia coli,Bacillus subtilis were purified respectively through a combination of ion-exchange and hydrophobic interaction chromatography.Specific activities of the CGTase from these three sources were 201 U/mg,200 U/mg and 199 U/mg,respectively.Chemical modification was further performed to improve the thermal stability of α-CGTase.The α-CGTase expressed in E.coli was cross-linked into a stable macromoleculs by glutaraldehyde.After water bathed at 50 ℃ for 30 min the stability of α-CGTase was improved as the cross-linked α-CGTase exhibit 78% left enzyme activity,which was 3.9 folds higher than that of dissociated α-CGTase.

Abstract:The extraction parameters of aminopeptidase by Ultra-flo system from the fermentation broth is studied in this manuscript.The optimum conditions for high high enzyme activity recovery and membrane flux were listed as follows: 30 kDa MWCO polyether sulfone(PES) membrane,the operation pressure between 0.2 MPa and 0.25 MPa,the initial pH value 5.5~6.5 and the concentration ratiof 3 times;The elution is in the form of regular flow of water,and the elution volume is about 10 times of the concentration of the fermentation broth.By the optimun conditions,the enzyme activity recovery rate was achieved at 85% and 79% of the desalination process,and the salt in the concentration of the fermentation broth has been removed about 90%,and the final concentration factor can be controlled for 8~10times.The enzyme activity recovery rate can be 67.15% of the whole of the Ultrafiltration process,and the membrane flux can be maintained above 16 L/(m2·h) throughout the process.

Abstract:A Box-Behnken design was used to optimize the extraction conditions of water-soluble polysaccharides(POP) from Polygonatum odoratum(Mill.) Druce.Three factors such as extraction time(h),extraction temperature(℃) and ratio of water to raw material were investigated.The experimental data were fitted to a second-order polynomial equation using multiple regression analysis and also examined using the appropriate statistical methods.The optimum extraction conditions were listed as follows:extraction time 2.24 h,temperature 88.35 ℃,and ratio of water to raw material 35∶1 mL/g,the predicted yield of polysaccharides extracted was 8.47%.Under the optimum conditions,the mean extraction yield of polysaccharides was(8.46±0.02).The antioxidant in vitro results showed that the inhibition effects of POP on 2,2-diphenyl-2-picrylhydrazyl free radical,hydroxyl free radical and superoxide anion radical with IC50 values were 0.646,10.029,0.241 mg/mL,respectively,which were higher than 0.048,0.238,0.078 mg/mL of ascorbic acid,respectively.POP showed significant inhibitory effects on hydrogen peroxide with IC50 value of 0.541 mg/mL,which was lower than 0.901 mg/mL of ascorbic acid.

Abstract:In this manuscript,the extraction technologies was optimized and the antioxidant activities of the total flavonoids from edible Gnaphlium affine was investigated.The microwave-assisted extraction technique of flavones from Gnaphlium affine and in vitro antioxidant activities of its flavones had been studied.The antioxidant activities of flavones were also compared with ascorbic acid(VC).The result shows that the optimum extraction conditions were:50% ethanol,1∶60(g:mL) material-liquid ratio,extraction temperature 60 ℃,and extraction time 6 min when the microwave power is 500 W.The average yield of flavones was 4.63%.Total antioxidant activities,scavenging superoxide anion and DPPH radical of the flavones from Gnaphlium affine were weaker than those of VC,but higher scavenging hydroxyl and restraining peroxidation of lipoprotein of yolk which induced by Fe2+.

Abstract:In this manuscript,the antioxidant activity(the reducing power,the ·OH and DPPH· eliminate ability) of a combined fungus polysaccharide extracted from a healthy oral liquid was studied in vitro and compared with raw materials.The results demonstrated that the antioxidant activity of all fungi polysaccharides showed stable growth along with the increasing concentration in the experiment.With the same concentration,the antioxidant activity of the polysaccharide compounds was higher than that of the polysaccharides extracted from Dictyophoraindusiataand CordycepsCephalosporium Mycelia,but was lower than or equal to the activity of lentinan.Therefor the combined fungus polysaccharide has greater health care function when consider the effect and the cost meanwhile.

Abstract:The effects of high-intensity pulsed electric fields in different levels of electric field intensity(20~35 kV/cm) and treatment time(100~400 μs) on microbial inactivation and antioxidant activity of strawberry juice were investigated in this manuscript.The changes of antioxidant activity of strawberry juice immediately after pulsed electric fields and conventional thermal processing(95 ℃,30 s) treatments and during the storage were further investigated.The results showed that compared with the conventional thermal pasteurization methods,the antioxidant activity of strawberry juice was less affected initially after PEF processing and were maintained in higher quality throughout storage period.

Abstract:In this study,air dried asparagus was ultrasonic extracted with 75 %(v/v)ethanol.The immune function of this extract on mice were investigated.Mice were orally administrated with asparagus extract at three dose levels(7,70,140 mg/Kg BW) for 30 days.At the organ level,thymus and spleen were weighed and their indices were calculated.The macrophage phagocytosis ability in vitro,transformation of spleen lyphmocytes,and serum hemolysin were investigated to assess the immune functions.The results showed that the middle and high dosage of asparagus extract promoted the spleen index and the phagocytic function of murine peritoneal macrophages.the middle and high dosage of asparagus extract significantly enhanced the transformation of spleen lyphmocytes and serum hemolysin(HC50).This suggest that asparagus extract can enhance both the specific and non-specific immune functions in normal mice.Moreover,the specific immune effects of asparagus extract especially depended on the dosage.

Abstract:The objective of this study is to investigate the feasibility and the effective does scope of hydrolysed casein bound iron(Fe2+-Cas hydr) on cure of iron deficiency anemia(IDA) with Ferrous glycinate(Fe2+-Gly) as the control.The IDA model for rat was first established,and then several groups were divided: the the experimental groups with Fe2+-Gly and Fe2+-Cas hydr in high,medium and low doses respectively,the control group with conventional diet and the Fe-deficient diet group.After 36 days,the weight and the hematology index were measured to analyze the effect of each treatment to anemia.It was found that the hematology index of rats in experimental groups with high and medium doses of Fe2+-Cas hydr recovered to normal range,and no significant differences appeared in the treatment effect of Fe2+-Cas hydr and Fe2+-Gly in the same doses.Fe2+-Cas hydr with iron ranging from 1.818 μg/(g.d) to 0.363 μg/(g.d) is most effective in treating anemia.
