Abstract:In recent years,since the special properties of the cyclodextrin(CDs),it has won more and more favors from different industries,such as food,cosmetic,pharmaceutical and so on.The CDs were in a great demand and also consumed with a stable speed around the world.As the most important factor in cyclodextrin industry,the cyclodextrin glycosyltransferase(CGTase,EC 2.4.1.19) became one of the hot points in CDs investigations.Today,less systematic work which related to the screening of CGTase strains had been done besides the catalysis mechanism and product specificity.This paper plans to directly describe the comprehensive means of obtaining the superior CGTase based on the future appeal of CD industry.Simultaneously,combine the former research results and the advantageous resources of our own country,in order to put forward expect of this field

Abstract:Providing balanced energy and nutrients for microbiota growth is esential for the maintenance of the colon ecosystem,and dietary fiber(DF) fermentation,particularly butyrogenic fermentation,augments colon health.Colon cancer,nflammatory bowel disease and other colonic disorders have become serious health issues.The effect of cereal DF,mainly nonstarch cereal polysaccharides and resistant starch on colonic microbiota,fermentation products of SCFA,microbiota modulation,as well as on colon health will be summarized.

Abstract:With the serious water pollution,the "algal bloom","red tide" formed by the overpopulation of algal is getting worse.At present,different methods including chemical,physical,biological were used to inhibit the growth of algae in polluted water.However,the characteristics of these methods are difficult controlling,high cost,and damaging the ecology.In order to develop a low cost and ecologically safe method to inhibit the growth of algae,plant allelopathy is introduced.In this review,the research progress of algal-inhibition with plant allelopathy was introduced,the types of plant allelopathy and the chemical composition of allelochemicals were summarized,the mechanisms and perspective of plant allelopathy were discussed.

Abstract:Cashew is one of the foods which can cause people allergic.In this manuscript,the sequence characteristics,secondary and tertiary structure,antigenic epitopes,hydrophobicities and surface accessibility,flexibility of allergen Ana o 2 from cashew were predicted and analyzed by using bioinformatics methods,such as Pubmed,SOPMA,swiss-model and DNAStar.Results demonstrated that the epitopes of Ana o 2 are probably at 108-111,181-186,217-218,234-238,244-255,283-287.These should be helpful for the further research of cashew allergen and be useful to develop of low sensitization cashew products.

Abstract:Effect of the different pH control strategies on the PSA production by Escherichia coli CCTCC M208088 was compared in this study.Those strategies including:(1)when 20 g/L K2HPO4 was presented in the medium,the titer of PSA achieved at 1.9 g/L PSA,however,the high-level residual K2HPO4 in broth bring difficult to the following PSA purification.(2)with 2.5 g/L K2HPO4 and feeding of 2 mol/L NaOH solution,the titer of PSA was 2.3 g/L;(3)the PSA titer was increased to 3.2 g/L by feeding of ammonia water as pH control agent.Furthermore,the feeding of ammonia could accelerated sorbitol consummation;(4) To further promote PSA production,a novel strategy using ammonia water for pH control coupling with sorbitol supplementation at a constant feeding speed was adopted.As a result,PSA production and productivity were increased separately to 4.8 g/L and 0.16 g/(L·h),which was 152% and 188% higher than that of the control(without pH control).

Abstract:In this manuscript,a novel T vector-mediated PCR technique was developed by using restriction enzyme digestion,pUCm-T vector ligation and nested PCR technique.This novel method can be used to determine two flanking unknown DNA sequences of a partial known DNA sequence.And this method was applied to clone the 5′-and 3′-end regulatory sequences of β-mannanase gene(Aus man5A) from Aspergillus usamii E001.The results showed that this method exhibited many advantages,for example,simplicity of operation,high specificity of primers,rapidness of result verification,and few restrictions for operation.By means of T vector-mediated PCR amplification,molecular cloning and DNA sequencing,the flanking regulatory sequences of the Aus man5A was successfully determined.

Abstract:The purpose of this manuscript is to study the inhibitory effect of the pomegranate peel polyphenols on the peoxidation of poly unsaturated fatty acid and to identify the key composition.For this,three classical anti-lipid peroxidation model,namely yolk system,low density lipoprotein system and rat liver homogenate system were used to investigate the effect of the purified pomegranate peel polyphenols(PPPs),Punicalagin and ellagic acid(EA)on the antioxidant activity.Results showed that PPPs exhibited a good protection against metal ions-induced lipid peroxidation in vitro in a dose-depend s.In addition,the trend in anti-lipid peroxidation activity was Punicalagin>EA>PPPs,from this we can concluded that Punicalagin the key anti-lipid peroxidation component in pomegranate peel polyphenol mixtures.

Abstract:In order to assess the potential of polyelectrolyte complex films composed of chitosan and sodium cellulose sulfate(NaCS) for controlled drug release,effects of chitosan Mw,Mw and substitution degree of NaCS on the mechanical(strength and brittleness) and swelling properties of the complex film were studied.The results showed that chitosan Mw,and the Mw and substitution degree of NaCS have great influence on the mechanical and swelling properties of chitosan/NaCS films.Higher elongation at break of the complex films would be obtained with chitosan of higher Mw.Meanwhile,the increase of the Mw and substitution degree of NaCS would cause the decrease of the swelling ratios.In addition,pH of the solution has great influence on the swelling ratios of chitosan/NaCS films.

Abstract:Ethyl carbamate(EC) is usually found in Chinese rice wine.It’s very important to directly remove EC from Chinese rice wine.Among various absorptive materials,two specific resins(L2,L3) were selected and mixed for effectively removing EC at a suitable addition of 10%(v/v).The removal rate was higher than 60%,meeting the EC limitation requirement while maintaining most of the components in Chinese rice wine.The removal of EC absorbed heat.And the temperature should be kept higher than 20 ℃ when conducted the removal.These results provided an applicable and convinient approach for directly reduce EC content in Chinese rice wine.

Abstract:The changes of microbial counts、acid and peroxide value duration of storage.With the accelerated shelf test,the study found out that the chief influencing factor of shelf life was the AV value.The TP could reduce the POV value and the AV value by 23.09% and 26.47% respectively.The changes of oil content and colour parameters were studied.Reduce oil content with coating materials,like sodium alginate and CMC-Na etc.The reduction of oil content and the change of colour parameters were studied.The sodium alginate had the best effect on the reduce oil content.It can reduce the oil content by 14.94%.

Abstract:In order to isolate a high lovastatin-producing Monascus strains,protoplast fusion technology was introduced in this manuscript.The protoplasts of Aspergillus terreus and Monascus anka was prepared under 5 hour and 3.5 hour enzymolysis,respectively,by mixed enzymes(0.5% lysozyme +0.3% Cellulase +0.3% snail enzyme) and then inactive by ultraviolet irradiation(20 W,30 cm)for 5 min and 4 min,respectively and mixed with PEG(30% PEG,0.05mol/L CaCl2,0.05 mol/L glycine).27 high lovastatin-producing strains were obtained from 187 fusants.Seven of them exhibited more than twice the yield of the original Monascus strain.

Abstract:The algl gene(GenBank accession number: GU585575) encoding alginate lyase(ALGL) from Bacillus cereus F1-5-10 was cloned into the vector pET-30a and expressed in Escherichia coli BL21 in this study.The optimum expression conditions of algl in E.coli was study and listed as follows: the concentration of IPTG 0.4 mmol/L,expression time 4 h,and expression temperature 32 ℃.Under the optimum conditions,the activity of recombinant enzyme achieved at 11.7 U/mg.Then,the recombinant enzyme was purified and the characteristics were determined.It was found that(1) the molecular weight was approximately 45 000;(2)the optimum pH and temperature were 6.6 and 35 ℃,respectively;(3)the activity was stimulated by 1.0 mmol/L Ca2+ and Mg2+;(4)the Km and Vmax values for alginate were 1.89 mg/mL and 15.01 U/mL,respectively.

Abstract:The objective of this study is to investigate the anti-fatigue activity of flavonoids from herbahouttuynia(FFH).For this,128 male mice were divided into control group,low-dose FFH treated group,medium-dose FFH treated group and high-dose FFH treated group.The control group was given dH2O and the treatment groups were given FFH(40,80、160 mg/kg) for 28 days.Body weight,blood lactic acid(BLA),serum blood urea nitrogen(BUN),tissue glycogen(liver and glycogen) concentrations in mice were determined.The results demonstrated that FFH extended the exhaustive exercise time of mice,effectively delayed the increasing of blood lactic acid,increasing the storage of tissue glycogen.From the above results,it can draw the conclusion that FFH exhibited significant anti-fatigue activity

Abstract:To improve the flavonoids yield from Ginkgo biloba leaves,the combination of microwave and high pressure technique was employed to extract the flavonoids,which were isolated and purified by macroporous resins.On the basis of single factor experiment,Box-Behnken combination menthod was used to study the effect of the three factors(pH,microwave extraction time and pressure) on the flavonoids yield and the optimum conditions listed as follows: microwave extraction time 2.58 min,276.62 MPa pressure,9.68 pH and solid-liquid 30 mL∶1 g.Under the optimal condition,the yield rate of flavonoids compound was 5.14%.By comparing the two factors of adsorptive capacity and desorption of the two adsorption resins,results showed that: AB-8 was the optimal resin of the two kinds of macroporous resins to separate the flavones from Ginkgo biloba flavonoids.The static adsorptive capacity,adsorptive capability rate and desorption rate were 16.2 mg/g,81.1% and 80.5%,respectively.After purification,the purified flavonoids mass fraction was 29.3%.

Abstract:The structure-function relationship of the protease from the fruits of siraitia grosvenorii was studied by analyzing the effects of modification on the protease activity.The activities were detected after the protease was modified by 8 kinds of protein modification reagents,respectively.And the results showed that the enzyme activity was significantly decreased after modified by PMSF,NBS and DEPC,but the others did not had obviously effected.So the hydroxyl group,indolyl group and imidazole group were demonstrated to be essential to the enzyme’s function probably,but the sulfhydryl group,amino group,garboxyl group,guanidino group and disulfide may not be essential for the activity of the enzyme.

Abstract:Polysialic acid(PSA) is considered an ideal alternative material of the polyethylene glycol(PEG) for protein or peptide modification due to its more effective safety in therapeutics.This research was a preliminary study of modification of CuZn superoxide dismutase(CuZn-SOD) with polysialic acid.The optimal conditions for the modifying reaction were as follows: PSA was activated by periodate oxidation,then coupled to CuZn-SOD in the PSA: SOD mole ratios of 40: 1 for 24h.It was shown that polysialylated macromolecule contained 3.9±0.3 molecules of PSA per molecule of SOD.SDS-PAGE electrophoresis and Atomic Force Microscopy(AFM) revealed that the molecular weight of polysialylated SOD was about 90~100kDa and the average size was 10~15nm,about four-fold of native enzyme size.Besides,it showed two kinds of microscopic spherical shape: coated single particle or coated multiple particles.As compared with native enzyme,the stabilities of modified enzyme against heat,acid,alkali and proteinases such as pepsin and trypsin were improved significantly.

Abstract:With the kelp as raw material,2% glutaraldehyde as pre-treatment agent,the optimum process parameters for the soaked kelp by Ultrasound microwave were determined in this manuscript.The optimum conditions were listed as follows: Ultrasonic power 50 W,microwave power 500 W,the processing time 60 s,liquid ratio 1∶20,extraction times 2.The optimal extraction conditions in assimilating action were obtained: pH 12,temperature 60 ℃,reaction time 2.5 h,liquid ratio 1∶20.Under the optimum conditions,the extraction rate for sodium alginate was 88.6% and the viscosity was 3670 mPa·S.
