    • Biosensor for β-Lactam Antibiotics Residue Detection-A Review

      Abstract:β-Lactam antibiotics residue trace detection has proved to be an important safety problem,due to its relationship of animal derived food safety and international trade.Biosensor which would rapidly and automatically detectingβ-Lactam antibiotics in convenient,has attracted more and more researchers.Based on the different biological reaction,it can be devided into three classes:enzymic,penicillin binding proteins(PBPs) and immune.Firstly,enzymic biosensor has been took into practice,but their capacities were not enough to residue detection.PBPs were a kind ofβ-Lactam antibiotics sensitive bacteria functional protein,which needs to be determined, characterized,and produced through advanced biotechnology.Comparative advantage of immunosensors was its low detection limit and systematic development.Recently,more research concerned with the immunosensors and studied on the new transducer,nano-technology and multi-residue automatically detection.New achievements and methodologies of computer, molecular biology and bioinformatics had been used to design and screen bio-recognition elements for biosensors development,as well as to investigate the biosensing process.It hopes that biosensor practicability would be improved though new biological research methods and findings.

    • Study on the Processing Stability of Mulberry Pigment

      Abstract:The red pigment from mulberry pigment was dissolved in the solution of 1,0.1和0.01mol/L hydrochloric acid,citric acid,tartaric acid,respectively.The pigment solutions were then heated at 70℃for 5 hours,processed ultrasonically for 1 hour and placed in the light naturally for 3 days,respectively.With the changes of absorbance(512 nm) as index to evulate the stability of pigment.The result showed that the pigment stability was affected by the kind and concent of acid solution.It was found that the pigment exhibited high stability in hydrochloric acid,but big changes of absorbance in tartaric acid and citric acid solution.

      Abstract:A variety of anticancer drugs induced cell death in the major form of cell apoptosis.It is of great significance to study the mechanisms on cell apoptosis for improvement the efficacy and development of new antineoplastic agents.Yeast is the single cell eukaryote that can be induced to undergo apoptosis,and mechanism of apoptosis in cells from yeast to mammals,are highly conserved,and that the yeast has become an important research model organism.This review focused on yeast apoptosis induced by the cytotoxic anticancer drugs,including the typical characteristics of apoptosis,key regulators,signal pathway,and apoptosis-related phenotypes.

      Abstract:The scavenging of·OH and·O2- by aqueous extractions from Various organs (inflorescence,leaf,aerial stem,rhizome,root) of Anoectochilus roxburghii(Wall.)Lindl were estimated.The results demonstrated that the aqueous extraction had capacity of scavenging·OH and·O2-.The cleaning effect on·OH was sorted as:leaf>rhizome、inflorescence>root>aerial stem.The cleaning effect on·O2- was sorted as:leaf、aerial stem>inflorescence>rhizome、root.

    • Influence of Oxygen Concentration on the Oxidation of Peanut Oil

      Abstract:The oxidation of fat and oils leads to the increase of peroxide value(PV) and flavor deterioration.More and more attention has been paid to prevent the oxidation of oils,especially in the case of consumer pack edible oils for its long shelf life.The purpose of this study was to investigate the effect of the oxygen content on the oil oxidation rate and flavor in airtight packages.Seven samples injected with different oxygen concentrations in the headspace of the erlenmeyers had been put in a 70℃oven to accelerate the oxidation.The results showed that the influence of oxygen concentration on the oxidation was significant.The PV of the sample with 51.8 mg injected oxygen was increased by 12.42 mmol/kg in 4 days,while only 3.58 mmol/kg of PV was increased for the sample with 3.2 mg oxygen injected.There was positive correlation between oxygen concentration an PV.

      Abstract:This study is to develop an efficient process for the lotus root juice by blanching with Color fixation.And the optimum color fixation compositions was determined and listed as follows:0.3%zinc acetate,0.18%disodium ethylenediamine tetraacetic acid and 0.02% ascorbic acid.Then the optimum formulation of lotus root juice was identified by the fuzzy mathematics Integrated Evaluation;lotus root purified juice 90%,White granulated sugar 8%, Citric acid 0.10%and Honey 0.10%.

    • Study on the Reduction of Lipid Peroxidization by Chitosan

      Abstract:The in vitro and in vivo effect of chitosan on the reduction of lipid peroxidization was studied in this manuscript.The results showed that chitosan at an addition of 0.02%had antioxidation effect on the lard and crude rape oil,but the activity was worse than that of ascorbic acid.0.05%chitosan and ascorbic acid exhibited the similar activity on the lipid peroxidization. And the chitosan significantly reduced the concentrations of serum free fatty acid and Malondialdehyde and but increased the activities of superoxide dismutase,catalase and glutathione peroxidase.Those results demonstated that chitosan decreased the lipid peroxidization through regulating the anti-oxidation enzyme activities.

    • Inhibiting Effect of Gingko Flavonoid on Lipid Peroxidation

      Abstract:Lipid peroxidation inhibiting activity of two types of Gingko flavonoid(i.e.aglycone and glycoside) was evaluated in this study.With the addition of the aglycone and the glycoside flavonoid into the soybean oil and the mice brain homogenate,peroxide value and the maleic dialdehyde(MDA) quantity were determined,respectively.Results showed that both the aglycone and glycoside flavonoids had efficiently inhibition effect on the lipid peroxidation,and aglycone exhibited stronger inhibiting activity(p<0.05).The aglycone is a more useful form of the Gingko flavonoid as the antioxidant additive.

      Abstract:In this study,the optimum conditions of gelatin and PVP to remove tannin in Blueberry juice were determined by orthogonal experiment.The optimum conditions for gelatin to adsorb tannin was listed as follows:adsorption temperature 30℃,adsorption time 30min,the dosage of 1%gelatin solution 10 mL/dL.The optimum conditions for PVP was as follows:adsorption temperature 30℃,adsorption time 25 min,thedosage of PVP 5 g/L.By employed those optimum conditions,the tannin removal rate achieved at 65.60%and 74.48%,respectively.

      Abstract:To meet the demand of fresh water fish process,this study choose the processing techniques of shred fish made by shred fish producer as the research model.The techniques parameters,such as striking time,digestion time,weight of fish and natural cooling time were studied by single factor experiment and Orthogonal experiment with sensory evaluation and fraction void as indexes.By the optimum techniques parameters,a high product efficiency was achieved.The results proven that a high production efficiency,simple manipulation and high product yield process was developed.

      Abstract:In order to improve the value of Chinese water chestnut and to reduce the potential pollution,acidolysis extraction of Pectin from Chinese water chestnut peel was investigated in this study.Back-Propagation neural network was used to optimize the extraction process.And the optimum conditions listed as follows:hydrochloric acid concentration was 0.81 mol/L, temperature was 78℃,ratios solvent to material was 19.5:1(mL:g),extracting time was 1.53 h.Under the optimum conditions,the yield of pectin reached at 11.76%.

      Abstract:By optimizing the experiment,mainly in Lycium Barbarum juice with seasoning after a mixed deployment,so that Lycium Barbarum juice mixture to achieve a better sense of the requirements of the senses in the Lycium Barbarum juice quality standards,based on the principle of fuzzy mathematics to establish the evaluation model,filter out the best formula.The results indicated that Lycium Barbarum powder flavoring agent for the best combination of A1B2C2 when the dosage of additives were suger14%,Kang Su-sweet 0.2%,citric acid 0.5%.

      Abstract:With the aim of increase the quality of flaxseed,SLG67-18.5 twin-screw extruder was used to investigate and optimize the effect of diameter of mould,crushed granularity, temperature,moisture content,rotation speed of screw and feeding speed on removal rate of HCN in flaxseed and extrusion intensity.Mathematic expressions of removal rate of HCN,in vitro protein digestibility and mould pressure was set up by quadratic regressive currency rotary design.And the optimum conditions were listed as follows temperature,moisture content, rotation speed of screw and feeding speed 156℃,16.6%,219 r/min and 76.1kg/h respectively. The results presented here provided knowledge to the development and use of flaxseed,and to the operation and adjustment of the existing extruders.

      Abstract:This paper studies the antibacterial action of Nisin and potassium sorbate compound in concentrated duck soup,inhibit spores and other heat-resistant bacteria in the soup by adding Nisin,which reduce sterilization intensity;TBHQ,BHA and BHT were added in the soup to research the anti-oxidant effect.The results showed that:when the concentration of Nisin and potassium sorbate were 0.20 g/kg and 0.03 g/kg,sterilization conditions reduced to 115℃for 30 min;The best compound scheme of anti-oxidant is:0.15 g/kg TBHQ+0.05 g/kg BHA.

      Abstract:This study investigated the detection methods and the corresponding detection parameters of paraquat(PQ).A serials of results were achieved:(1)The minimum limition of HPLC was 12 mg/L,the recovery ration was 42~52%,the RSD was 4.43%,the precision was very high.(2) the parameters of ELISA were:minimum limitation 0.005μg /L,the recovery ratio 70~80%,RSD 8.15%;.(3)The detection results of GICA is very good,minimum limition 50 mg/L,through a standard color reflected series.Among these detection protocols,GICA method has been proven to be a simple,efficient,quick method to determine the content of PQ in food.

      Abstract:Coupling nano-probe technology,aptamer recognition with resonance light scattering analysis,through recognizing reaction between adenosine and its specific aptamer in homogeneous matrix,causing the disaggregation of the aggregated network structure of adenosine specific aptamer functionalized nanogold structure,a sharply change on the resonance light scattering signal can be obtained.Based on it,a novel homogeneous aptamer recognizing based method for adenosine detection was established.The linear concentration range was 0.1~10μmol/L for adenosine with a detection limit of 0.04μmol/L(3S/N).The proposed method also shows good selectivity for adenosine detection and no interference from guanosine,thymidine,cytidine and uridine.

    • An Efficient Method for Glutathione Extraction from Dried Saccharomyces Cells

      Abstract:An efficient method for glutathione(GSH) extraction from the dried Sacchromyces cells was proposed in this study.During extraction at(70±1)℃,the mixture of acetic acid-hydrochloric acid was used to adjusted pH 3.5,the ratio of solid to liquid(w/v) was kept at 1:8. GSH extraction result reached at 5.5 mg/g,which was 54.49%-62.24%higher than that of all of the reported extraction process.After treatment through micro-filtration(0.45、0.22μm) and ultra-filtration(5 kD MWCO),60.98%of the yeast protein was removed.The collection from ultrafiltration was analyzed by high performance liquid chromatography and amino acid analyzer, the result showed that glutamic acid was the major portion in amino acids impurity besides protein and small peptide in crude extract,and these would provide basic data for purifying GSH.

      Abstract:In this study,yeast invertase(β-fructofuranoside hydrolase) was extracted and purified by cell disruption,heat denaturation,organic solvent precipitation and DEAE-Sepharose Fast Flow chromatography.Five chemical modification agents(PMSF,DTT,SUAN,NBS,EDTA) and several other metal ions were used to determine the amino acid residues involved in the active site of yeast invertase.The result indicated that serine residues and metal ions were inessential to the invertase activity while the lysine and cysteine residues were linked to the enzyme activity, which were not in the active site of the enzyme.That the indolyl ring of tryptophan residues could be modified by N-bromosuccinimide and these groups could be protected by sucrose revealed that the tryptophan residues were present at the invertase active site.

      Abstract:This study use 11°P different brands of beer as model to investigate the effect of iso-αacid and protein content on the beer foam stability.The beer foam stability,iso-αacid content, the concentration of foam protein and protein Z and lipid transfer protein(LTP) content were determined by the national standard method,reversed-phase high performance liquid chromatography(RP-HPLC method),bicinchoninic acid method(BCA method) and sodium dodecyl sulfate Sodium - polyacrylamide gel electrophoresis(SDS-PAGE method).The results showed that in the iso-co-humulus which was part of iso-αacids and the total protein content have more closely relationship with the foam stability,but the content of a single protein Z,or LTP holding of less closely with the foam stability.This research provides an alternative methods to improve the foam retention time.

    • Study on the Technology of High-Acidity Rice Vinegar Submerged Fermentation

      Abstract:In this manuscrpt,a novel step-start protocol of fermentation process was developed to inhibit the bacteria aged and to short the fermentation starting-up time.By compared with the oxygen transfer efficiency in the clear solution and unfiltered solution,it was found that the mash-solid of unfiltered solution exhibit a negative effect on the oxygen transfer efficiency.By combinatorial of multi-stage fermentation process and semi-continuous fermentation,a serial of results were achieved:the acetic acid productivity increased from 0.12 g/dL·h to 0.19 g/dL·h;The fermentation time reduced 4 hours;The final acidity of high-acidity vinegar semi-continuous fermentation was more than 9 g/dL,the productivity reached at 0.18 g/dL·h, higher 20%than that of the similar studies.

      Abstract:This study was carried out to explore a feed protein production from the corn bran by mixed- stain solid-state fermentation with Trichoderma koningii and Candida tropicalis.With the single-factor and orthogonal experimental optimization of the nutritional and environmental conditions for xylanase activity and protein production,the optimal conditions were as follows: wheat bran 25%,solid-liquid ratio 1:1.5,pH 5.5.Under the above conditions,the true protein content in the final product achieved>20%after the fermentation at 30℃for 7 d,and the xylanase activity reached 1 300 IU/g,which diplayed a considerable potential to future practical application.

    • Soft Sensor Modeling of Burkholderia cepacia Lipase Fermentation Process Based on BP Neural Network

      Abstract:The aim of this study was to establish a soft sensor model of the Burkholderia cepacia lipase fermentation process.Procedure and method:for this,the backpropagation neural network was applied to rebuild a soft sensor model for the fermentation process of Burkholderia cepacia lipase,and genetic algorithms was further employed to optimize the initial weights and thresholds of neural networks,so as to speed up the convergence rate and achieve a comprehensive optimum result.Result:The model could imitate precisely the process of bacteria growth,culture medium consumption and lipase production,which suggests that the model possesses good reliability and generalization ability.The backpropagation neural network combined with genetic algorithms is an effective and quick method for simulating and predicating fermentation process of B.cepacia lipase.

    • Comparison of Mycelial Growth Kinetics Model for Pleurotus ostreatus Cultured in Shaking Flask and Fermenter

      Abstract:This manuscript study the difference of the fermentation kinetics of Pieurotus ostreatus TD300 in shask and 40 L fermentor.It was found that the mycelial growth kinetics models of both culture modes were accord with Logistic equation.Culturing period,maximum specific growth rate and mycelial concentration cultured in shaking flask were 8 d,0.050 h-1 and 1.5001 g/ml,respectively.While the corresponding value changed to 84 h,0.110 h-1 and 1.4750 g/ml when Pieurotus ostreatus TD300 cultured in fermenter,respectively.The results proven that fermentor culutre was a higher efficiency culture for Pieurotus ostreatus.Furthermore,pellet size and number per unite volume of liquid spawn cultured by shaking flask were smaller and higher than that by fermenter.It was probably caused by the viscosity of medium in shaking flask reduced faster than that in fermentor.

    • Research on the Purication of Polysialic Acid Based on the Property of the Fermentation Broth

      Abstract:In order to obtain Polysialic acid(PSA) from the fermentation broth,the property of PSA and methods to remove the impurities in the fermentation broth were studied in this manuscript.The results demonstrated that PSA showed a good heat-stability and anti-alkalescence capability,and it was precipitated by ethanol and cetyl pyridinium chloride(CPC) in certain condition;impurities in the fermentation broth can be removed by filtration with Perlite as filter aid.Based on this,a novel PSA purification process was established.Through this process, the final PSA product was obtained with a purity and recovery(98.1±1.6)%and(56.1±1.7)%,respectively.

    • Isolation and Identification of Cholesterol-Lowering Lactic Acid Bacteria from Spinach

      Abstract:Total 33 strains of Lactic acid bacteria were isolsted from spinach,using four different kinds of media.All strains could reduce cholesterol from media by sulfate-phosphate-ferric methods,and the percentage of cholesterol-removal rate ranged from 4.82%to 47.58%.There are 9 strains exhibited the percentage of cholesterol-removal above 40%,among them,the cholesterol-removal percentage of strain MB65 achieved at 47.58%.Based on morphological, physiological,biochemistry test and 16S rDNA sequencing analysis,strain MB65 was identified as Lactobacillus casei.

    • Fermentation Conditions Optimization and Kinetics Analysis for Cell Wall Polysaccharides Production of Saccharomyces cerevisiae in Batch Culture

      Abstract:In order to further increase the production of the cell wall polysaccharides from Saccharomyces cerevisiae and to better understanding of the physiological properties of cell growth,products formation and substrate consumption,the environmental conditions of Saccharomyces cerevisiae were optimized in 5L fermentor.Firstly,the effect of stirrer speed,pH value and temperature on biomass,glucan and mannan production were carefully investigated by single factor experiment,and the optimum fermentation conditions were determined as follows: stirrer speed 400 r/min,pH value 4,temperature 32℃.Experimental models were constructed using BP neural network to train experimental data and the BP models and fitting curves were determined.The predicted values of the models were compared with the experimental values. These models were proved to well simulate the change of biomass,sugar consumption and cell wall polysaccharides production.

    • Identification of Lepista sordida LS7 and Antibiotic Activity Analysis of its Fermentation Broth

      Abstract:A strain LS7 was isolated from the Laoshan mountain of Qingdao.According to the morphological,cultural characteristics,and ITS and 5.8s rDNA sequence analysis,the isolate was identified as Lepista sordida.The fermentation broth was extracted by different organic reagents,and their antibiotic activity was also evulated in this study.This strain exhibited the inhibitory action on Gram-positive bacteria,Gram-negative bacteria and fungi,especially for Fusarium oxysporum and Colletotrichum capsicibutl.The results shown here provided the theoretical basis for the application of Lepista sordida in food and medicine industry.

    • Improvement of Extracellular Laccase Production by Paecilomyces sp. S152

      Abstract:The culture medium containing maltose(20 g/L),bean peptone(10 g/L) and yeast extract(1 g/L) was found to favor laccase production by Paecilomyces sp.S152 with an enzyme activity of 5064 U/L.The intra- and extra- cellular location of laccases were detected in the cells of Paecilomyces sp.S152.To improve cell permeability,Tween 80(1.5 g/L) was added to culture broth and an enhanced extracellular laccase activity was detected with the shortened fermentation period(6 days) and improved(946 U/(L·d).
