      Abstract:The success of licensed phosphodiesterase type-5 (PDE-5) inhibitor drugs (viz sildenafil,vardenafil and tadalafil) has led to the widespread use as adulterants in herbal dietary supplements or herbal medicines.Which cause a threat to the public health,in this review,the apply progress of TLC,HPLC,LC-MS and MECC in the analysis of known PDE-5 drugs with the standards were summarized.However,only LC-ESI-MS/MS can be competent for the identification of both the licensed PDE-5 drugs and unknown analogues in herbal products simultaneously.It is believed that Immunoassay may be a promising strategy for rapid detecting of undecared PDE-5 drugs in a simple,sensitive and high-throughput way.

      Abstract:4-α-glucanotransferase has the function of generating large-ring cyclodextrin.The crude enzymes production by the genetic mutant of E.coli DH-5α-TA (CGMCC No.3093)was separated and purified by high-temperature processing (65℃,20 min),Ni-NTA affinity column and dialysis.The purified enzyme was demonstrated by SDS-PAGE to be a homogeneous protein and the molecular weight was estimated as 57KDa.It was proved to be with the high level of transglycosylation activity by HPLC which was always used to measure the oligosaccharide and its smallest substrate was maltose.The enzyme exhibited high stability at the temperature range from 70℃ to 85℃ and the pH range from 6.0 to 8.5.The optimal temperature for the enzyme was 75℃ and optimal pH was 7.5,respectively.

      Abstract:The quality changes and hygroscopic capacity of carrot chips dried by different drying process (hot-air drying,vacuum microwave drying,freeze drying and combined freeze-vacuum microwave drying) were investigated in this manuscript.The quality parameters such as VC and carotene contents,color,expansion ratio and drying time from different drying process were compared.it was found that freeze drying exhibit advantages on VC and carotene contents,but its expansion ratio much better than that of freeze drying,reduced drying time and hygroscopic capacity was less than those treated by freeze drying.Compared with that of freeze drying alone,the combined freeze-vacuum microwave significantly reduced drying time and hygroscopic capacity.

      Abstract:As is a special starch modality,possessing granule quality but not crystalline,Non-crystal granular corn starch has preferable enzymatic degradation,reactivity and adsorption property.Bake-drying with washing with ethanol,vacuum bake-drying,atmospheric bake-drying,vacuum freeze-drying and spray-drying were applied to dry non-crystal granular corn starch for scale-up production.It showed that different drying technology affected morphology of non-crystal starch,which was analyzed scientifically.

      Abstract:The target of this manuscript is to elucidate the mechanism of TPS reducing blood glucose,for this,the scavenging effect of 89% and 51% tea polysaccharides (TPS) on hydroxyl radical,superoxide radical key carbohydrate metabolic enzymes activities,and the glucose transportation to 3T3-L1 were careful investigated in vitro Results showed that the abilities of scanvenging hydroxyl radical and superoxide radical of TPS with lower purity was abvious,while,that with higher purity was not obvious.The effect on α-glucosidase activities and transportation of glucose to 3T3-L1 cell were also no obvious.However,TPS significantly enhanced the activities of hexokinase(HK) and glucokinase (GK).

      Abstract:In order to overcome the shortcomings of the methods including water-drying,salt-drying and vacuum freeze-drying,this study firstly prepared the silver nanoparticles by chemical reduction in liquid phase,than separated the corruption bacteria from the sea cucumber,.Based on the above results,the sterilizing effect of the silver nanoparticles on the corruption bacteria and on the all microorganisms in the putrid sea cucumber were carefully investigated.It was demonstrated that 0.05 mg/L silver nanoparticles can efficiently sterilize 80% bacterium after 12 h treatment.

      Abstract:This manuscript is to study the membrane contamination removal technology process in the process of preparating food grade soy lecithin by inorganic membrane micro-filtration.Through comparing the effect of n-hexane washing and NaOH solution washing on membrane fiux and the acetone insoluble content,it was found that n-hexane with low moisture efficient delay the membrane fouling,and the highest membrane flux can be acquired with washing by 2 g/dL NaOH solution in 80℃ and 2 hours.The acetone insoluble content also arrived at the highest value (more than 60%).

      Abstract:Our research were focused on physical,gelatinized and retrograded properties of starches from twenty broad bean varieties in China in order to provide a theory foundation on their processing suitability.We determined several physical parameters,the viscosity variety under up-and down-temperatures,the strength of broad bean gels and cooling stability of broad bean starch systems.The physical,gelatinized and retrograded properties of starches from twenty broad bean varieties in China were distinctly different with different cultivated areas or varieties.The average WBC of starches from twenty broad bean varieties was 97.97 %.Swelling power and solubility of broad bean starches increased markedly with increasing temperature over 50 ℃.The highest freeze-thaw syneresis ratio and light transmittance of broad beans starches were YNFD and QJXLC,respectively.The onset gelatinized temperature of these broad bean starches was 60.25~73.08 ℃.The highest peak viscosities,holding strength,setback,gel strength of broad beans starches were BSDBC,ZJQPCD,DBHQC,CLCD,respectively.The cooling syneresis ratio of TX2 starch gel was the lowest among all broad bean starches.

    • Antitumor Effect of Lycopene Nanodispersion on Mice Transplanted with Hepatoma

      Abstract:To investigate the effect and mechanism of lycopene nanodispersion on tumor growth,lycopene nanodispersion was prepared by the emulsification-evaporation process (the ultrasonic emulsification).The antitumor effect,antioxidant activity and anti-proliferation activity in vivo were studied with the model of mice transplanted with hepatoma H22.The results showed that lycopene nanodispersion was obtained by the process.When mice transplanted with hepatoma H22 were treated with 1.95 mg/kg and 3.9 mg/kg lycopene nanodispersion for 10 days,the tumor growth inhibitory rate were 32.37% and 54.34% respectively.The intake of lycopene nanodispersion led to the increase of glutathione peroxidase (GSH-Px) activity,and the reduction of the mice serum malondialdehyde (MDA) content.It also led to the increase of total antioxidant capability (T-AOC) and superoxide dismutase (SOD) activity,but decrease the content of MDA and hydrogen peroxide (H2O2 ) of liver of mice.It also could inhibit the expression of proliferating cell nuclear antigen (PCNA) in tumor tissue.These results suggested that lycopene nanodispersion can significantly suppress the growth of hepatoma H22 in vivo.The increases of anti-oxidation activity and anti-proliferation activity may contribute to the antitumor mechanism.

    • Effects of Taurine on Egg Production,Immune Responses and Fat Metabolism in Laying Quails

      Abstract:With the quails as research model,different levels of taurine (0,0.01% and 0.05%) were added to the diets to carefully investigate the effects of taurine on production,immune responses and fat metabolism of laying quails.A total of 108 quails were allocated to 3 dietary treatments.Each treatment comprised 4 replicates of 9 quails.The experimental period lasted 4 weeks.The results showed that 0.01% taurine decreased broken and soft eggs ratio significantly (P<0.05),0.05% taurine increased laying rate,decreased feed convesion rate and broken and soft eggs ratio significantly compared with the control (P < 0.05).Compared with that of the control,0.01% taurine decreased triglyceride and atherogenic index significantly (P < 0.05);0.05% taurine significantly increased serum HDL-C and LPL lipoprotein lipase and significantly decreased triglyceride and atherogenic index (P < 0.05).Results of this study indicated that taurine can increase laying rate and decrease broken and soft eggs ratio in laying quails,taurine can enhance immune function,and taurine can promote fat metabolism through increasing serum HDL-C level and LPL lipoprotein lipase activity in laying quails.

    • Lipase-catalyzed Synthesis of L-ascorbyl Stearate in Non-aqueous Phase

      Abstract:In this manuscript,the technical conditions of Novozym 435 lipase-catalyzed synthesis of L-ascorbyl stearate (L-AS) in non-aqueous medium were determined with 20% lipase and 1:2 L-ascorbic acid to stearic acid (molar ratio).The optimum reaction conditions listed as follows:30ml t-butanol with 6.82mmol L-ascorbic acid,13.64mmol stearic acid,0.24g lipase and 3.0g 4A molecular sieves,shaker speed 150r/min,the reaction carried out at 55℃ water bath with 48h.Through the separation and purification,the isolated product was finally identified as L-ascorbyl stearate by infrared (IR) and mass spectrometry (MS) detection,the highest yield reached 64.7%.Lipase reused five times,the lowest yield was 61%.

    • Study on the Quality of Jucy Peaches during Store Using an Electronic Nose

      Abstract:The aroma composition in jucy peaches was detected by an electronic nose during diferent storage time.The aroma composition emanating from the Jucy peaches was sampled by PEN3 systems,and the response values of PEN3 was obtained,The data was analyzed using principal component analysis (PCA) and linear diserimination analysis (LDA),respectively.The results showed that electronic nose was able to better classify the jucy peaches by LDA method,and a clear distinction was available among jucy peaches on 0day,day1~day3 and day4~day6.Moreover,The changes of aroma composition reflected the regularity of physiological and quality change in jucy peaches during the storage time.The sensor 6,sensor8 and sensor5 played important roles in the quality detection of Jucy peaches by using loadings analysis.

      Abstract:The antimicrobial properties of Compound film of Chitosan and bamboo vinegar were studied in this manuscript.The optimum combination of liquid mixtures with 0.6% bamboo vinegar and 1.0 g/dL chitosan was firstly determined by evulating thethe minimum concentration (MIC) and the inhibitory activity of the film.Compound preservations were made with chitosan and bamboo vinegar mixture as main reagents and glycerol as the auxiliary.The water solutions of the compound preservations with different concentrations were filmed on the surface of the bracken,and then preserved at room temperature.The results showed that the coating antistaling agent contained compound preservations 1.5% and glycerin 0.6%.Kept the bracken three time marinated into the coating antistaling agent at 30 ℃ temperature.The preserved bracken mainly kept its original taste and nutritious value,and remained green without mildew.

    • Optimization of the Extraction of Polysaccharides with Microwave-Assisted Extraction from Actinidia chinensis

      Abstract:Microwave-assisted extraction (MAE) was applied to the polysaccharides extraction from the dried root,stem and leaf of Actinidia Chinensis.Three key operation parameters,including the extraction time,the power of microwave and the ratio of material to solvent,were optimized with uniform experimental design method.The optimal conditions for extracting three raw materials were determined by the regression equations.Considering the efficiency and the feasibility of experiment,the actual operating conditions were 0.016 9 of the ratio of material to solvent,24 min of the extraction time and 385 W of the microwave power in the extraction of root;0.0170 of the ratio of material to solvent,23.8 min of the extraction time and 539 w of the microwave power in the extraction of stem;and 0.016 1 of the ratio of material to solvent,23.5 min of the extraction time and 539 W of the microwave power in the extraction of leaf,respectively.The extracted yields of polysaccharides in the actual optimal conditions were 7.0%,3.7%和3.2%.The deviation between the values from experiments and calculation from root and leaf were both below 5%,which demonstrated that the regression equation could predict the yield of polysaccharide in the given conditions well.

      Abstract:N-maleyl chitosan oligosaccharide (NMCOS) and N-phthaloyl chitosan oligosaccharide (NPCOS) with the same substituting degrees (0.25) were synthesized.The structure were characterized by FTIR spectra,the molecular weight was measured by Gel Permeation Chromatog-raphy (GPC).Their antioxidant activity were evaluated by the scavenging of hydroxyl radical (.OH) and 1,1-Diphenyl-2-picrylhydrazyl radical ( DPPH).The results showed NPCOS have stronger hydroxyl and DPPH radical scavenging activity than that of NMCOS.The results presented here demonstrated that the antioxidant activity of N-acyl Chitosan Oligosaccharide with the same substituting degrees was determined by the substituting groups.

      Abstract:In this study,Chaoshan fish sauce was firstly prepared by incubating the mixtures of anchovies at different salt contents (10%,15%,20%,30%)and koji contents (0%,5%) conditions for 30 days.Then further increase the salt contents to 30% for 180 days fermentation.The changes of indexes,including total soluble nitrogen(TSN),amino-acid nitrogen (AA-N),total volatile base nitrogen (TVB-N),pH value,non-enzymatic browning index,free amino acid composition,chromatic aberration and the sensory evaluation were determined during 210 days fermentation.The results demonstrated that both low salt content and low salt content integrating of koji were advantage to the fish sauce fermentation.Those two fermentation processes increased the contents of TSN,AA-N and free amino acid (Glu,Ala,Lys),when compared with the corresponding indexes of the traditional Chaoshan fish sauce.And the optimum conditions were 15% salt with 5% koji.

    • Isolation and Identification of a Tacrolimus Producing Strain

      Abstract:In order to isolate a tacrolimus producing strain,soil samples were initially treated with 75 μg/mL K2Cr2O7 and 2 μg/mL benzylpenicillin.Seventy-one actinomycetes were isolated by anti-fungal activity against Candida albicans then the metabolites origin from those isolations were further analysis by HPLC.Among these,only one tacrolimus producing strain was determined.It was identified by culture characteristics,morphological characteristics,physiological,biochemical properties and 16S rDNA sequence and phylogenetic analysis.The screened strain was gram-positive,the growth temperature ranged from 15 ℃ to 38 ℃(the optimum was 28 ℃);the growth pH ranged from 6.5 to 8.5(the optimum at 7.0).The strain can grown on glucose,sucrose,maltose,xylose,fructose,lactose and rhamnose.Based on the results of the general taxonomical characteristics and the 16S rDNA sequence,the isolated has proven to be Streptomyces and named Streptomyces sp.NJYH2618.

      Abstract:The target of this study is to develop a efficient extract process of inulinase from Aspergillus ficuum by solid state fermentation.For this,the extract process parameters were carefully investigated and listed as follows:0.1 mol/L sodium acetate buffer of pH 4.5 was used for extraction solvent,the ratio of solid to liquid of 1:20.5,temperature of 39.6 C,stirring rate of 207 r/min,extraction time of 40 min.By the optimum conditions,the enzyme recovery yield was increased by more than 50%,when compared with that of the initial conditions.

    • Identification and Phylogenesis Analysis of Cultivable Microorganism Isolated from Fresh Hami-Melon Juice

      Abstract:Forty-eight strains were isolated from the fresh hami melon juice using the dilution plate method.Those isolated strains divided into ten groups according to the morphological features.Then the 16S rDNA sequences were performed by employing the universal prime bacterial for polymerase chain reaction (PCR) amplification.The alignment search was performed with BLAST and GenBank databases,then compared this sequences using the software of Clastal1.81,the phylogenetic tree was built using Mega 4.1 software.From the phylogenetic tree,it could be concluded that Bacillus subtilis is the main cultivated bacteria in the fresh hami melon juice and the following is Bacillus licheniformis,Bacillus mojavensis,Pseudomonas aeruginosa strain RE7,Burkholderia cepacia strain ATCC 21809,Acinetobacter baumannii,and Enterococcus sp,Klebsiella sp.

      Abstract:Recycling the stilliage condensates produced into the fermentation phase,coupled with the alcohol fermentation and the anaerobic biogas fermentation.It has been demonstrated that acetic acid and propionic acid exhibited main inhibition effect on the alcohol fermentation.The concentration of acetic acid inhibit alcohol fermentation was 0.4 g/dL and that of propionic acid was 0.1 g/dL.When the lactic acid concentration increase to 2.5 g/dL,a inhibition effect on alcohol process was detected.

      Abstract:The immobilization conditions,and application acid urease via adsorption and embedded were investigated in this study.The optimum concentration of sodium alginate,calcium chloride and chitosan was 2.0,5.0 and 3.0 g/dL,respectively.the optimal immobilized time was 2.5h,the immobilization rate was 82.3%,and the reaction take place at 40℃and pH4.The half-lives of immobilized acid urease was 48 days,the unit of immobilized urease was 3.6U/mg,the urea removal rate was 65% at 35℃,24 h and the addition level of immobilized acid urease was 0.1g/mL (the quality of immobilized enzyme /the volume of Chinese rice wine),under the same conditions,the urea removal rate still reached at 39.7% under the continuous using thirteen times.

      Abstract:Screening fine fermentation strains in Lactobacillus bulgaricus and Streptococcus thermophilus for goat′s yogurt.The fermentation characteristics of the four strains Lactobacillus bulgaricus and four strains Streptococcus thermophilus were studied during goat yoghurt manufacture.The results showed that Lb-800 strain has higher producing acid rate,weaker postacidification and abundant alive bacteria within four strains Lactobacillus bulgaricus.St-499 strain has higher producing acid rate,higher producing viscosity ability and abundant alive bacteria within four strains Streptococcus thermophilus.The goat’s yoghurt has suitable acidity and viscosity,no whey separates and the number of alive lactic acid bacteria more than 1×109cfu/mL,while the ratio of fine strains Lb-800 and St-499 is within 2:1~1:2.

      Abstract:The effect of the nutrition and environmental conditions,including dissolved oxygen,pH,added sugar and other factors on Natamycin production by Streptomyce natalensis SY-4795 were careful investigated,and the optimum conditions listed as follows:the dissolved oxygen 30%~35%,the constant sugar concentration in fermentation broth keep at 2%,pH 5.2~5.4.With the optimum fermentation conditions,natamycin production increase 3.01 g/L to 5.32 g/L in batch fermentation.

      Abstract:A bacteria strain was isolated and identified for L-lactic acid production in this study,The isolated strain was identified as Bacillus coagulans based on the results of 16S rDNA,the physiological characteristics and the biochemical properties.Then the optimum nutrient conditions was determined by the single factor experiments and list as fellows:10.5 g/dL corn syrup,1 g/dL yeast extract,1 g/dL cotton-seed protein.Furthermore,it was found that the presence of Mn2+、Fe2+ and Mg2+ in the medium can enhance L-lactic acid production.

      Abstract:In order to further improve the titer and yield of neokestose production by Xanthophyllomyces dendrorhous,the medium compositions and environmental conditions for neokestose production were optimum by the single factor experiment and the orthogonal experiments,and listed as follows:sucrose 110 g/L,dried corn steep liquor powder 9 g/L,pH 6.5,rotation speed of shaker 220 r/min and inoculation size10% in 25 mL of culture medium in the 250 mL flasks at 18 ℃ for 24 h.The concentration of neokestose by Xanthophyllomyces dendrorhous 269 was achieved at 73.2 g/L,the yield of neokestose on sucrose reached at 66.5%,which was 1.87 times than that of the control.

      Abstract:In this manuscript,response surface design was used to optimize the nutritional and environmental conditions of cellulase production by Penicillium glaucum NS16.There are 18 factors affect the cellulose production,including the fermentation time,shaking rate,concentration of carbon sources,nitrogen sources and inorganic salts.The influence of the 18 factors was evaluated by Plackett-Burman design.The result showed that wheat bran,CMC-Na and the fermentation time play important roles on the cellulase production.According to the result of Plackett-Burman design experiment,the value range of the significant influential factors could be estimated,and a Box-Behnken design was employed to optimize these factors and the results were shown in response surface graph.Following the results,the optimum conditions listed as follows:6.0 g/L wheat bran,7.0 g/L CMC-Na,and 96 hours feimentation time.By the optimum conditions,309 IU/ml of cellulase was produced by Penicillium glaucum NS16,which was higher 227% than that of the control (94.51 IU/mL)

      Abstract:The purpose of this manuscript is to screen the marine microorganisms and evaluate the antagonistic effects of those isolated microorganism to Escherichia coli and the Staphylococcus.A isolated marine microorganism L having better antagonism effects was separated from the diluted marine mud.Physiology and biochemical characteristics of the isolated marine microorganism,the curve of fermentation control parameters were studied.Antagonism effects of the ferment production were tested with the Escherichia coli and the Staphylococcus.The result shows that marine microorganism L was marine actinomycetes,the suitable fermentation time is 9 to10 days,the secondary metabolite in fermentation broth have certainty the antagonism ability to Escherichia coli and Staphylococcus.

    • Pilot Study on Effects of Semen strychni Fermented by Six Different Fungi on Toxicity,Analgesia and Anti-Inflammation

      Abstract:The toxicity,the analgesic and anti-inflammatory effect of Semen strychni integrative fermentation with six different fungi,including Schizphylhls commne Fr.,Shiraia bambusocola P.Henn,Coriolus unicolor (L.:Fr.) Pat.,Cantharellus Cibarius Fr.,Beauveria bassiana (Bals.) Vuill and Cordyceps gunnii (Berk.) Berk were evaluated in this manuscript.When Semen strychni raw materials (RMSS) were solid-fermented with those fungi,the corresponding product were named as SCSS,SBSS,CUSS,CCSS,BBSS and CGSS,respectively.LD50 (i.g.) in mice were determined by the Bliss method to evaluate the acute toxicity of RMSS,processed Semen strychni (PSS) by traditional Chinese method and the six fermented products.The body writhing induced by i.p.0.1% acetic acid and the ear edema induced by 2% croton oil were used to test the analgesic and anti-inflammatory effects of six fermented Semen strychni products with the positive controls of diclofenac sodium (DSE) and RMSS.The toxicities of SCSS,SBSS,CUSS,CCSS,BBSS and CGSS were lower than that of RMSS,and were similar to that of PSS.The analgesic and anti-inflammatory effects of the aboved six fermented products were similar to that of RMSS.In conclusion the six different fermented Semen strychni had the same advantageous effects as that of PSS and the acute toxicity of them could be decreased compare to that of RMSS.
