      Abstract:In this manuscript,the enzymatic hydrolysis of rice straw in three different forms: original(untreated),pretreated by dilute acid,and pretreated by a sequence of dilute acid and dilute alkali,were studied.The hydrolysis was carried out in an enzyme/substrate ratio of 20 FPU/g,8%(w/v) substrate concentration,45 ℃ for 72 h.The digestibility results suggested that the cellulose hydrolysis was greatly affected by the presence of hemicellulose and/or lignin in the rice straw.The cellulose conversion ratio 43.4% from dilute acid-treated rice straw was 2.6 times higher than that 16.8% from untreated rice straw,whereas from dilute acid and alkali rice straw such value was higher 3.6 times(60.6%).This result was probably due to the fact that the strong modification in the material structure caused by the hemicellulose and lignin removal from the rice straw.As a consequence,the cellulose fibers were separated being more susceptible to the enzymatic attack.It was concluded that the lower the hemicellulose and lignin in the rice straw,the higher efficiency of cellulose hydrolysis could be achieved at.

      Abstract:The polysaccharide composition from Hippophae rhamnoides L.polysaccharideⅠ(HRPⅠ) was study in this manuscript.Two different compositions,HRPⅠa,HRPⅠb were found in the isolated crude polysaccharide by DEAE-Sephadex-50 column the compositions of HRPⅠa contained xylose,mannose,glucose in 1∶1.06∶1.13 molar ratio.Furthermore,HRPⅠa configuration is composed of α and β-glycoside.

      Abstract:The antisepsis effect of Nipagin esters on the fish tidbit was carefully investigated in this manuscript,Compare with that of traditional preservative-potassium sorbate,the inhibition effect of E.coli was observed and the best result of the microorganism growth when propylparaben,Butyl p-Hydroxybenzoate,n-heptyl p-hydroxybenzoate was mixed at the ratio 1:1:1 and the gross of all kinds of Nipagin esters is 0.2 of the fish tidbit.

    • Comparative Analysis of Flavor Compounds in Draft Soy Sauce Origin from Two Different Membranes

      Abstract:In this manuscript,two different membranes,namely organic membrane and inorganic membrane were used for raw-soy sauce filtration to obtain draft soy sauce.Solid phase micro-extraction(SPME) and GC-MS were applied to analyze the flavor compounds of the draft soy sauce.Totally 70 compounds were identified from those two samples.These compounds included alcohols(18 kinds),phenols(5 kinds),esters(7 kinds),aldehydes(13 kinds),ketones(5 kinds),acids(2 kinds),heterocyclics(8 kinds) and hydrocarbon(6 kinds).Among of them,the main flavor compounds were alcohols,phenols,aldehydes,ketones heterocyclic.The results of this study provided evidence for the improvement and enhancement of the traditional fermentation of soy sauce.

      Abstract:In this study,the physiological parameters of melatonin live,including inflammatory cytokine IL-1,IL-10 of spleen,intestinal myeloperoxidase(MPO) were determined after LPS treat.It was found that content of IL-1 and MPO activity decreased but IL-10 increased after 4 h stimulation by LPS.For the case of 16 h,the values of IL-1,IL-10 and MPO activity were lower than that of 4 h.The results indicated melatonin can depress inflammation induced by LPS,maintain inflammatory and anti-inflammatory balance and protect animals from injury.

      Abstract:The aim of this study is to develop a method on the process of microwave freeze drying abalone,nano-silver coating was introduced and the technique experiments were carried out.The results was showed that nano-silver of 0.3mg/L coating could sterilize about 99.1% and the count is less than 30 of Coliform bacteria on the process of microwave freeze drying.To ensure perfect sterilization effect,nano-silver coating technique was combined with microwave freeze drying process.Nano-silver coating treatment could lead to low microorganism number and had no significant effect on drying efficiency.

      Abstract:In this manuscript,the determination of optimum conditions of the rapeseed protein hydrolization was performed and the results listed as follows: hydrolyization time 5 hours with alcalase at pH 9.5,55 ℃,3% substrate and enzyme concentration 5500 u/g,the hydrolysis rate could reach to 21.14%,then hydrolization with flavourzyme for 3 hours at pH 6,50 ℃ and enzyme concentration 900 u/g,the bitter value of rapeseed protein hydrolysates was very low under such conditions.The results of decoloration experiment of rapeseed protein hydrolysates with active carbon showed that the optimum conditions were: active carbon dosage 1.5%,pH 4.5,temperature 55 ℃,and reaction time 50 min,the decoloration rate could reach to 32.15%,and the amino acid loss rate was 25.15% under such conditions.

      Abstract:The extraction technology of polysaccharides from blueberry after extracting anthocyanin by cellulase and ultrasonic was studied in this manuscript.Also,decolorizate rough polysccharides and remove protein,then through column chromatography,finally test the antioxidant capability of the refined polysccharides SP.the optimum conditions for process was determined by Orthogonal test experiment and listed as follows: time 100 min,temperature 40 ℃and enzyme additive 0.6%,the extraction percent 2.319%.In the extraction process by ultrasonic,the optimum conditions were: the extraction time 40min,temperature 50 ℃,power 80 W,the extraction percent 2.335%.Compared with that of the traditional extraction process by water,the process suggested in this manuscript has been proven easier manipulation and get higher extraction yield.Furthermore,the results also strong demonstrated that SPⅠhas stronger antioxidant activity.

      Abstract:The objective of this study is to investigate scavenging activity of tetramethylpyrazine(TMP),an effective constituent isolated from the rhizome of nature medicinal plant Ligusticum chuanxiong Hort.,on free radicals in vitro and inhibitory effect of TMP on H2O2-induced oxidative damage of endothelial cells.The inhibition of H2O2-induced intracellular reactive oxygen species(ROS) by TMP was analyzed on a FACS Vantage SE flow cytometer.The suppressive effect of TMP on nitric oxide(NO) production was measured by Griess method.The scavenging effect of TMP on free radicals including hydroxyl radical(·OH),hydrogen peroxide(H2O2),DPPH free radicle(DPPH·) and lipid peroxidation free radical was determined by colorimetry method.It was found that TMP inhibited H2O2-induced production of intracellular ROS and exhibited strong scavenging activity against hydroxyl radical.In addition,TMP displayed scavenging activity against DPPH·,NO,H2O2 and lipid peroxidation.The results presented here indicated that TMP may exhibit scavenging activity on free radicals in vitro and inhibitory effect on oxidative damage of endothelial cells.

      Abstract:The thermal properties of D-ribose,L-cysteine and Maillard reaction of non-aqueous phase ribose/cysteine model system were detailed investigated through thermal analysis methods such as thermal gravity,simultaneous differential thermal analysis and differential scanning calorimetry.The thermal behavior of the formation of volatiles in the reaction of ribose/cysteine was also studied by thermal gravity-mass spectrometry.It showed that the degradation temperature of L-arabinose and cysteine,respectively,were higher than that of their mixed counterparts.The earlier reaction of L-arabinose/cysteine was an endothermonic process and it must be depend on the reaction temperature and reaction duration under specific temperature.Most of volatile compounds produced in the second phase of weight loss and hydrogen sulfide also occurred in this period.It shown that Strecker degradation of cysteine into hydrogen sulfide required more higher reaction temperature.Furthermore,sulfurous compounds could generate as a result of the formation of hydrogen sulfide.Hence,the formation of sulfurous compounds had strong temperature dependence.

      Abstract:The manuscript described the extraction of the category SOD-L in hawthorn leaves and fruit.This experiment extract and analysis is through using pyrogallol since the oxidation of hawthorn in leaves and fruit category SOD-L,we measured extract of SOD-L vigor in hawthorn leaves and fruit was 630.14 u/mg and 252.05 u/mg,respectively.SOD-L in the bran has a typical absorption at 280 nm.And it was found that the activity of SOD increase with the presence of Mg2+,Cu2+.SOD-Lactivity of hawthorn in leaves and fruit in the bran had an obvious optimum stability at the condition of 40 ℃ and pH 8.0.

      Abstract:The subject mainly through optimizing anti-extraction program,study the feasibility of the technology of extracting organic acid from anaerobic acid mixture by supported liquid membranes.The experiments showed the supported liquid membranes system which composed as the following: hydrophobic polypropylene hollow fiber membrane,the extractive solvent,the organic solvent and the anti-extractive solvent can be efficiency extracted organic acid.The best condition in the system is that: extraction reaction time is 360 minters,NaHCO3 in the anti-extraction is 20 g/L,pH is 9,concentrate ratio is 2.0.The technology of extracting and concentrating organic acid from anaerobic acid mixture by supported liquid membranes can be preferably realized.

      Abstract:The obective of this study is to isolate mangrove forest derived actinomyces strains for obtaining the bioactive metabolite producing strains to investigate antifungal and anti-tumor active metabolites.For this,actinomyces strains were isolated by single colony isolation technique.The antifungal activity was assayed by paper method.The anti-tumor activity was assayed by MTT method.Ninety-five strains of actinomyces were isolated from 3 soil samples and 2 water samples collected from a mangrove forest in Futian,south of Shenzhen,China,of which 94% were streptomycetaceae.Nine pathogens were used to test the antifungal activity of the ninety-five strains,among of them 31.6% of the fermentation broth showed the antifungal activity.Two tumor cell lines(A549 and K562) were used to test the antitumor activity with the MTT method.The result indicated that 21% of the fermentation broth presented the antitumor activity.Furthermore,the fermentation broth of the s33,s62,s65and s68 showed antifungal activity toward more than five pathogens and antitumor activity on the two cell lines.

      Abstract:The effect of pH shock on the performance of reactor for simultaneous anaerobic sulfide and nitrate removal was investigated in this study.The response of pH in reaction system to the pH shock was very sensitive,even though there were phosphate and carbonate to buffer pH variations.The practical pH responsive curve was different from the theoretical one caused by hydraulic retention time distribution,which means that is the comprehensive results from physical,chemical and biological interaction.The sulfide-oxidation was infuenced in some way by pH shock,but recoverable.The higher pH in reaction system,the lower sulfate concentration in the effluent,which suggested that high pH inhibits the sulfur oxidation.The nitrate reduction was sensitive to pH shock,but the responsive curve of effluent nitrate concentration slightly lagged behind,which was possibly related with the mechaniam of nitrate uptake.

      Abstract:In this study,the antitumor effect of the metabolites from Stigmatella WXNXJ-B was analyzing by HepG2 cell using MTT method.Furthermore,the stability of the metabolites under different physical and chemical environment was examined and the qualitative characteristics of the metabolites were analysis.The results showed that the antitumor bioactive metabolites efficient inhibited the growth of HepG2 cell.The optimum fermentation time of antitumor substance was 7 days.HPD100,XAD16 and DA201 resin were efficient to adsorb the bioactive metabolites.The antitumor bioactivity on HepG2 cell line did not obviously change when the temperature was lower 70 ℃,pH was in the range from 3 to 10 and the metabolites were treated with daylight and ultraviolet light during 12 h.It was detected that lactones,glycosides and steroids components existed in the metabolites.

      Abstract:Fluorescent-labeled molecules have been widely used in biological detection and diagnosis.In this manuscritpt,the Ru(bpy)32+-doped silica nanoparticles were firstly prepared with a modified W/O method.Human IgG(hIgG) was measured based on the specific interaction between hIgG and functionalized fluorescent core-shell nanoparticle-labeled gost anti-hIgG.The calibration graph for hIgG was linear over the range 1-100 ng/mL with a detection limit of 0.3 ng/mL.From 11 measurements,the R.S.D.is 2.2% for samples of 30 ng/ mL of hIgG.The results demonstrated that the method have a potential advantages of sensitivity,simplicity and good reproducibility for the determination of hIgG,and is applicable to the determination of hIgG in serum samples.

      Abstract:In this manuscript,the taste bud cells of fungiform papillae of ICR mice were select as research model and Percoll gradient centrifugation was used as purify method.The methods of trypan blue staining and immunohistochemistry staining were performed to compare the viability and purity of taste bud cells of before and after purification respectively.Taste bud cells were further identified by transmission electron microscopy.Results showed that taste bud cells,which were purified by discontinuous Percoll gradient centrifugation,were remarkably separated and mainly located interface between 20% and 40% Percoll working solution;the purity of taste bud cells were distinctly improved from less than 10% to more than 65%(p<0.01) by this treatment;the viability of taste bud cells were up to 94% after purification by trypan blue staining and the results of transmission electron microscopy further confirmed taste bud cells from ultrastructure.

      Abstract:Remote homology detection between protein sequences is one of the principal research objectives in structural and functional genomics.Proteins with similar structure and function but low sequence similarity consist of protein superfamily.Therefore,the detection of remote homologues is the task of identifying protein superfamily.In this manuscript,a clustering algorithm,called ModuleFind,based on network modularity was presented.The method maximizes the modularity of protein network to find the partitioning with strong community structure.The resulting algorithm gives high quality of clusters quantified by F-measure that combines precise and recall,in the experiments of the detection of the remote homologues based on the superfamily level of SCOP database.

      Abstract:NADH kinase is the critical pathway for NADP(H) biosynthesis in yeast.In Saccharomyces cerevisiae,mitochondrial NADPH is mainly supplied by a mitochondrial NADH kinase encoded by POS5 gene.However,a mitochondrial NADP+-specific isocitrate dehydrogenase encoded by IDP1 gene could also supply mitochondrial NADPH.Here,through the phenotypic study of POS5 single mutant,IDP1 single mutant,POS5IDP1 double mutant,including their growth ability,temperature sensitivity,non-fermentable carbon sources usage ability,amino acid synthetic ability and anti-oxidation,supplying mode of mitochondrial NADPH and function of NADH kinase in mitochondria were investigated.

      Abstract:Reconstruction of gene regulatory network can help to explore the essence of life.The Linear Combination Model has been successfully applied to the reconstruction of the gene regulator network,since it is very simple and can be easily solved.However,the linear relationships between genes were only considered in the model.In order to circumvent this problem,energy factor has been added in the Linear Combination Model,thus the new model can be used to analysis the nonlinear relationships between genes.Then the present model has been applied to reconstruct the gene regulatory network of Escherichia coli on SOS DNA repair process.Our result demonstrated that the proposed model can be reconstruct SOS DNA repair process well and also improve the accuracy.

    • Enhancement of 2-keto-L-gulonic production in Gluconobacter oxydans Through Feeding Metalions

      Abstract:To further increase the production of 2-keto-L-gulonic acid by Gluconobacter oxydans,the effects of six different metal ions were determined,based on the analysis of L-Sorbosone pathway.It was found that Fe3+,Mg2+,Mn2+ can enhance the cell growth and 2-KLG production.Then,an optimum concentration of those three metal ions was achieved by developing a multivariate quadratic equation: Fe3+ 0.21 mmol/L,Mn2+ 9.98 mmol/L,and Mg2+ 4.23 mmol/L.By using the optimum combination,the production of 2-keto-L-gulonic reached at 65.1 g/L,which is 144.4% higher than that of the control.

      Abstract:The target of this study is to achieve the cloning and expression of ricin A chain gene and produce the recombined ricin A chain protein(RTA) with high activity ane then assay it activity by RTB mediated by recombined adenovirus the RTA could enter cells and exert cytotoxicity.For this,the ricin A chain gene was amplified by PCR and cloned it into the fusion expression vector pET32a to construct plasmid(pET32a-His-RTA),the plasmid was transferred into E.coli BL21 and induced with low concentration of IPTG(1 mmol/L) at low temperature(20 ℃).Purified RTA by Ni-NTA column and the purified protein was identified by SDS-PAGE electrophoresis and Western-blot.It was found that the constructed recombinant plasmid(pET32a-His-RTA) was efficient expressed RTA fusion protein and about 50mg purified protein(Mr 47 000) was obtained from 1 liter culture broth.The cell death rate significantly increased and the highest being about 50-60 percentage point in the system containing RTB.

      Abstract:Mycobacterium sp.SH5 was a potential strain for degrading phytosterol.After mutation by UV and NTG,a mutant with 9α-hydroxylase defective was selected and named as Mycobacterium sp.SH5-52.On the basis of mutation,the nutritional and environmental conditions of Mycobacterium sp.SH5-52 were investigated and optimized.With the optimum conditions,Mycobacterium sp.SH5-52 could degrade 0.6% substance phytosterol to Androst-4-end-3,17-dione(AD) and Androst-1,4-end-3,17-dione(ADD) with the average conversion ratio significantly improved from 37.2% to 47.6%.

      Abstract:The nutritional and environmental conditions for acetoin production from Bacillus licheniformis in flasks were studied in this manuscript.Firstly,the environmental conditions,such as pH value,culture temperature,seed age,volume and inoculation ratio,and nutritional conditions,like carbon and nitrogen sources were carefully investigated.Based on the results of the single factor experiment,the orthogonal experiment was carried to get the optimum medium composition.Then fed-batch fermentation was performed with glucose feeding.The yield of acetoin achieved at 20 g/L in fed-batch fermentation with glucose feeding.
