Abstract:研究了高压脉冲电场(PEF)对乳铁蛋白(LF)的抑菌性能的影响。结果表明,LF质量浓度、电场强度、脉冲频率、脉冲数的增加,有利于LF抑菌能力的增强,而温度的升高会使之减弱。电场强度为35 kV/cm时,LF的抑菌能力达到最大值。LF的质量浓度越高,对电场强度的变化越敏感。当脉冲个数达到744时,LF的相对抑菌能力提高了34%。处理温度为15~55℃时,PEF处理的LF的相对抑菌性能增长了5%左右。当温度升高至65℃时,LF的相对抑菌能力下降显著,降低了约22%。

Abstract:采用正交试验和单因素试验相结合的方法对树莓叶片中精油的提取工艺进行了研究,得出的最佳工艺参数为:破碎60目;液料质量比为8∶1;浸泡14 h;水蒸汽蒸馏6 h。各因素对提取结果影响的程度依次为:蒸馏时间>破碎程度>液料比>浸泡时间。NaCl的加入量80 g/L可显著提高得油率。蒸馏速度200 mL/h(为每小时蒸出蒸馏器容积10%的馏出液)。

Abstract:对显齿蛇葡萄中二氢杨梅素的超声辅助溶剂提取工艺进行了研究,在单因素实验和正交实验的基础上,得出超声波法辅助溶剂提取显齿蛇葡萄中二氢杨梅素的最佳工艺条件为:在40℃下,以液料体积质量比为15 mL∶1 g的体积分数65%乙醇溶液,超声波辅助溶剂提取40 min,目标物二氢杨梅素的一次提取率可达93.1%,优于常规的热提取法。

Abstract:研究了采后1-甲基环丙烯(1-MCP)处理对绿芦笋贮藏品质的影响。结果表明,1-MCP处理可明显抑制绿芦笋贮藏过程中的呼吸作用;对VC和叶绿素含量的下降、丙二醛积累均具有抑制作用;提高了超氧化物歧化酶的活性,降低了过氧化物酶和苯丙氨酸转氨酶的活性,从而延缓了嫩茎衰老进程,对减少芦笋采后损失和保持品质,具有较好的作用。其贮藏期在室温下可延长2~3d,4℃下可延长5~10 d。

Abstract:分析比较了乌骨鸡与非药用鸡种鸡肉间的总脂质含量和脂肪酸组成的差异,为研究乌骨鸡总脂质的营养与补益功能提供基础。以相同条件养殖的非药用鸡种崇仁麻鸡、岭南黄鸡鸡肉为对照,测定3种鸡肉中的总脂质含量。采用GC-MS联用技术对3种鸡脂肪的脂肪酸组成进行定性、定量分析。结果发现:3种鸡肉中崇仁麻鸡的总脂质含量最高,乌骨鸡的含量最低。3种鸡的总饱和脂肪酸(SFA)和不饱和脂肪酸(UFA)差别不显著,乌骨鸡鸡肉中多不饱和脂肪酸(PUFA)、必需脂肪酸(EFA)和花生四烯酸质量分数分别为26.4%、23.38%、3.21%,显著高于其他两种鸡。乌骨鸡总脂质较非药用鸡种的总脂质有更好的营养价值。

Abstract:雪莲多糖是一种水溶性的功能性天然多糖,作者比较了料液比、温度、提取时间等因素对新疆雪莲组织培养物中雪莲多糖提取率的影响。结果表明,雪莲水溶性多糖的最佳提取工艺条件为料液比为1 g∶12 mL,温度85℃,时间2 h.粗多糖最大得率为7.08%。此粗多糖总糖质量分数为15.37%,从糖含量的角度,新疆雪莲组织培养物提取的粗多糖是野生雪莲提取的粗多糖的优良替代物品。

Abstract:研究了β-胡萝卜素氧化降解产物的生理活性。对利用OsO4为催化剂,将β-胡萝卜素在OsO4-H2O2-Ether体系中进行控制型氧化降解,对氧化降解产物进行薄层分离后,采用体外细胞培养技术观察β-胡萝卜素氧化降解产物对Hela细胞以及bel-7402细胞增殖的影响。结果发现,β-胡萝卜素氧化降解产物在高浓度时对Hela以及bel-7402的抑制作用强于β-胡萝卜素的,且对bel-7402的作用强于对Hela的,显示β-胡萝卜素氧化降解产物对癌细胞的增殖有抑制作用。

Abstract:为了研究产甘油假丝酵母高产甘油和耐高渗的机制,以Zeocin作为选择标记,考察和比较了两种转化方法,即醋酸锂法和电转化法的转化效果,以建立简单有效的产甘油假丝酵母转化体系。结果表明,细胞生长时期是影响转化率的关键因素。在OD600约为1.3时制备感受态细胞,在电压为1.5 kV时进行电击,可获得较高转化率,为每微克DNA 139个转化子。在OD600约为1.0时制备感受态细胞,醋酸锂预处理细胞1 h,获得的转化率为每微克DNA 154个转化子。综合考虑,醋酸锂法更适合于产甘油假丝酵母的转化。

Abstract:研究了脂肪酶固定化及其催化大豆油制备生物柴油的工艺。采用溶胶-凝胶法对脂肪酶进行了固定化,考察了固定化酶催化大豆油转酯化的生产工艺中酶用量、醇油比、含水量、反应温度、反应时间、溶剂等参数对转酯过程的影响。实验结果表明,当大豆油4.5 g时,最佳的反应条件为:固定化酶646 mg,醇油摩尔比4∶1,含水质量分数为6%,40℃,甲酯的最终转化率为96.33%。

Abstract:用木瓜蛋白酶对方格星虫(Sipunculus nudus)进行酶促水解,并对酶解物冻干粉的成分进行了分析,其中粗蛋白质量分数为69.02%,多肽质量分数为60.60%,肽的平均链长为3.73个氨基酸残基,肽分子的平均相对分子质量为447.6;游离氨基酸占5.27%。氨基酸分析结果显示,方格星虫酶解物含有17种氨基酸(色氨酸未测),其中人体必需氨基酸含量较高,占总氨基酸数的30.62%。方格星虫酶解物中还含有P、Fe、Mg、Mn、Zn、Cu、Se等至少12种矿质元素,以及含有抗氧化作用的营养成分,并通过动物试验证明了方格星虫酶解物具有显著的抗氧化作用。方格星虫酶解物可用于食品添加或用作功能性食品。

Abstract:确定了副溶血弧菌多克隆抗体的制备方法:用体积分数0.05%甲醛于30℃灭活副溶血弧菌4 h制备抗原,并将此抗原分多次,以不同剂量免疫新西兰大白兔获得特异性多克隆抗体,以山羊抗兔IgG-HRP作为酶标二抗,通过间接ELISA法测定其多克隆抗体的效价、敏感性和特异性。结果表明:在免疫的第5周产生大量高效价抗体,效价最高可达1.6×105;此多克隆抗体对副溶血弧菌检测灵敏度为1×105CFU/mL;交叉反应和阻断试验结果显示,此多克隆抗体具有很强的特异性。

Abstract:用响应面法对三孢布拉霉(Blakeslea trispora)液体发酵β-胡萝卜素的培养基进行了优化。首先用快速登高路径逼近β-胡萝卜素最大产量区域,然后根据快速登高法的实验结果进行响应面实验。运用逐步回归分析法,获得β-胡萝卜素产量与柠檬酸、棉籽油、黄豆粉的最优回归方程,且分析了各因子间的交互效应。最后,通过岭脊分析(Ridge max)得到β-胡萝卜素产量最大值时的组合为:柠檬酸3.10 g/dL、棉籽油11.28 g/dL、黄豆粉2.54 g/dL。β-胡萝卜素产量可达1.02 g/L,比优化前提高了25%。

Abstract:L-阿拉伯糖异构酶能将D-半乳糖异构成D-塔格糖。通过单因素试验和快速登高法对乳酸菌SK1.002产L-阿拉伯糖异构酶的培养基进行优化,确定发酵优化条件为(组分g/L):麦芽糊精28,酵母膏10,玉米粉浆22,无水乙酸钠10,K3PO40.2,NaCl 0.01,FeSO4.7H2O 0.01,Mg-SO4.7H2O 0.2,MnSO4.2H2O 0.05,L-阿拉伯糖2.5。发酵初始pH 8.4,培养温度37℃,接种体积分数3%,培养时间12 h。在此发酵条件下,酶活达到了7.28 U/mL。
