Abstract:采用流变学法测定了亚麻籽胶溶液的胶凝点、熔化点,并采用质构仪、扫描电镜和原子力显微镜等手段研究了影响亚麻籽胶凝胶强度的因素,结果表明亚麻籽胶具有胶凝性,它能形成一种热可逆的冷致凝胶,亚麻籽胶溶液的胶凝点低于其凝胶的熔化点,且亚麻籽胶溶液的胶凝点及其凝胶的熔化点均随冷却的起始温度的升高而升高。亚麻籽胶浓度、溶解温度、pH、NaCl、CaCl2及复合磷酸盐能影响亚麻籽胶的凝胶强度,亚麻籽胶的凝胶强度随着浓度的增加及溶解温度的升高而增强;在pH6~9的范围内,亚麻籽胶的凝胶强度达到最大;NaCl和复合磷酸盐可以降低亚麻籽胶的凝胶强度,低浓度(〈0.3%)的CaCl2可以增强亚麻籽胶的凝胶强度,而高质量分数(〉0.3%)的CaCl2能降低亚麻籽胶的凝胶强度。

Abstract:乙醛脱氢酶和乙醇脱氢酶是嗜热厌氧乙醇菌Thermoanaerobacter ethanolicus乙醇代谢途径中的关键酶。根据高同源性的NADP(H)依赖型Ⅱ型乙醇脱氢酶(S-ADH)的序列设计引物,从T.ethanolicusJW200基因组中PCR扩增出编码S-ADH的基因,插入带组氨酸标签的pET-20(b),测序获得基因大小为1 059 bp,并在大肠杆菌JM109(DE3)中表达。表达产物经Ni亲和柱提纯达电泳纯。带组氨酸标签的重组NADP(H)依赖型乙醇脱氢酶具乙醇脱氢酶和乙醛脱氢酶双活性,能将乙酰辅酶A转化成乙醇.带组氨酸标签的重组酶酶学性质为乙醛脱氢酶活性最适值为pH 8.4,最适温度70℃;乙醇脱氢酶活性正、逆反应分别最适pH 8.0,最适T 55℃;最适pH8.9,最适为T 60℃。以乙酰辅酶A(Aceyl-CoA)为底物,该酶的Km为3.32 mmol/L,Vmax为12.5μmol/(min.mg)。T.ethanolicusJW200中双活性S-ADH的首次发现,建立了该菌乙醇代谢途径中从乙酰辅酶A到的乙醇的另一条通路。

Abstract:通过正交设计等实验方法,详细的研究了培养基成分、酶系组成、菌龄、渗透压缓冲剂、pH、酶解温度、时间和再生培养方式等多种因素对Fusarium maire原生质体制备和再生的影响。实验结果表明:将Fusarium maire通过先静止2 d再以180 r/min震摇2 d后,以0.6 mol/L的KCl配制的pH为5的复合酶(3 mg/dL溶壁酶、2.5 mg/dL蜗牛酶1、mg/dL溶菌酶和0.5 mg/dL纤维素酶)中,32℃恒温酶解4 h,原生质体制备产量达3.2×107个/mL;纯化后的原生质体在25℃,以0.6 mol/L的蔗糖配制的YCM再生培养基上单层培养,其再生率达8.9%。为今后的通过原生质体融合和诱变研究,构建紫杉醇工程菌奠定了基础。

Abstract:用可降解淀粉微球吸附薄荷油制得了包合物,建立了包合物中薄荷油的快速测定方法——紫外分光光度法。测定了吸附时间和投油量对饱和吸附量的影响。结果表明:该方法快速、准确、重复性好、操作简便。吸附2h,薄荷油体积分数为4%时,饱和吸附量84.74μL/g淀粉微球。

Abstract:以体外抑制肿瘤细胞增殖的活性作为筛选导向,从灰树花深层发酵培养菌丝体中提取、分离纯化得到一酸性糖酞GFPS1b.HPGPC法测得其重均相对分子质量为21000.蛋白质质量分数16.60%,糖质量分数为81.32%。利用化学方法(单糖组成分析、部分酸水解、甲基化分析)和仪器方法(GC、GC—MS、红外吸收光谱、核磁共振)分析GFPS1b的糖链结构,结果表明其单糖组成主要有G1c、Ga1、Ara和少量的糖醛酸,其糖链主链由两个α-D—G1c(1→3)、α-D—Ga1(1→4)、α-D—G1c(1→3,6)和α-D—Ga1(1→6)组成的,其中葡萄糖基的6位发生取代,取代侧链主要为α-L—Ara-(1—4)-α-D—Glc(1→。可以认为GFPS1b为一新糖肽。

Abstract:在比较不同的提取方法后,选用乙醇为提取溶剂。通过单因素实验结合响应面法对白首乌中抗氧化成分的提取工艺进行优化研究,确定了提取工艺的最佳条件为温度:70.1℃,乙醇体积分数:75%,提取时间:1.78 h,提取次数为两次。在此条件下白首乌醇提取液的总抗氧化能力理论值为158.68 U/g,实测值为164.07U/g±3.73。

Abstract:对金丝小枣多糖生物活性进行了研究。经过鼠脾、鼠腹腔细胞增殖等试验表明,金丝小枣多糖与对照组相比,在30-200μg/mL剂量范围内显著地促进鼠脾淋巴细胞的增殖(P〈0.01),且显示出明显的剂量-活性关系(R^2=0.9489);金丝小枣多糖也可促进腹腔巨噬细胞的增殖,在30-100μg/mL剂量范围内,金丝小枣多糖与对照组相比差异显著(P〈0.05);金丝小枣多糖在体外有明显的抗补体活性。

Abstract:利用纤维素酶(EC 3.2.1.4)和蛋白酶(EC 3.4.14.9)从黑木耳当中提取出了2种多糖,相对分子质量分别为3.17×105和1.83×105。以正常小鼠和糖尿病小鼠为对象,对黑木耳多糖的降血糖功能进行了研究,使用剂量为100,200,400 mg/kg(以体重计)。结果表明,当质量分数在200 mg/kg以上,黑木耳多糖能够显著降低糖尿病小鼠的血糖值,但对正常小鼠的血糖值没有影响。黑木耳多糖能够增加糖尿病小鼠的糖耐量。
