Abstract:采用80℃热水提取,得到当归水溶性多糖W-ASP。经阴离子交换柱层析、凝胶过滤柱层析对其进行纯化分级。实验结果表明:W-ASP11主要由葡萄糖组成,相对分子质量约为380000;W-ASP12主要由半乳糖和阿拉伯糖组成,相对分子质量约为19000;W-ASP2和W-ASP3主要舍半乳糖、阿拉伯糖和鼠李糖以及少量葡萄糖和甘露糖,并含有较高的糖醛酸,由单糖组成及红外图谱初步分析,可判断是一种果胶类多糖。当归多糖主要组分W-ASP3经凝胶色谱(GPC)鉴定为均一组分,其相对分子质量约为62000。

Abstract:采用硫酸铵分级沉淀、透析脱盐、DEAE-Cellulose离子交换色谱等分离纯化技术,从Aspergillus ficuum菌株混合酶系中分离得到4种菊粉酶组分,应用薄层层析法分离各组分水解菊粉的产物,发现其中3种组分主要含外切菊粉酶,一种主要含有内切菊粉酶。进一步对所得到的内切菊粉酶组分酶解菊粉制备低聚果糖进行了研究,研究了底物浓度、加酶量、反应温度和反应pH对低聚果糖制备的影响,确定其最适反应条件为:底物浓度50g/L、加酶量10U/g底物,反应温度45℃,反应PH6.0。在此条件下反应72h,菊粉酶解率达74%,低聚果糖得率可达50%以上,酶解产物以DP2~DP4为主。

Abstract:研究了外源质粒DNA经胃肠道吸收可能对肝脏产生的作用机制。给Balb/c小鼠灌胃质粒pcDNA3 200μg,在灌胃后4h分离肝脏,提取肝脏的总RNA。利用寡核苷酸芯片对灌胃质粒pcDNA3后的Balb/c小鼠肝脏进行基因表达谱研究。结果发现17664个基因中,表达上调100条,表达下调41条。按基因功能分类,表达上调的基因可分为免疫应答基因、转录因子基因、信号转导基因、转运相关基因及代谢相关基因等;表达下调的基因主要为脂质代谢基因。灌胃外源质粒DNA后,肝脏组织主要表现为急性时相反应的加强、免疫反应的活化、细胞信号通路的活化以及脂质代谢途径的抑制。表明外源质粒DNA通过胃肠道途径可广泛调控肝脏的基因表达。

Abstract:用乙醇作为溶剂,采用常规浸提法及超声辅助法对元宝枫叶总黄酮的提取进行了对比研究。通过单因素及正交试验结果得到浸提法的优化条件为:溶剂为体积分数50%的乙醇溶液,料液比1g:25mL,提取时间1.5h,浸提温度70℃。按照此条件提取两次,第一次提取率94.16%,第二次1.36%;超声辅助法的优化条件为:溶剂为体积分数50%的乙醇溶液,料液比1g:15mL,提取时间0.5h,浸提温度60℃。按照此条件提取两次,第一次提取率为96.29%,第二次提取率为3.11%.

Abstract:作者研究了以德氏乳酸杆菌为生产茵种,以未添加酒花的麦汁作为培养基生产乳酸的主汁生物酸化技术,用于调节啤酒生产过程pH值。确定5^#乳酸菌的最适生长温度为42℃,最佳生物酸化麦汁质量分数为14%。研究表明,液体培养基的组成对该菌产乳酸有较大的影响。生物酸化麦汁调酸比约为1.40左右,不会对成品啤酒风味产生太大的影响。

Abstract:对桑果红色素的提取及稳定性进行了研究,结果表明:桑果红色素的最大吸收波长为480nm,较适宜的提取剂为乙醇,最佳的提取条件为:提取剂体积分数为80%,提取剂pH值为6,提取温度为40℃,提取时间为2h。该色素耐光、耐热、抗氧化、抗还原性强,常用的食品添加剂蔗糖、食用香精、维生素C以及质量浓度小于0.1g/dL的金属离子Mg^2+、Al^3+、Ca^2+、Cu^2+、Fe^3+等对色素无明显影响。

Abstract:通过对外源酶酶解枸杞效果的研究,结果表明:外源酶能显著提高枸杞浸提液中可溶性固形物和营养成分的含量,其单一纤维素酶和木瓜蛋白酶浸提枸杞的适宜pH值分别是5.0~5.5和pH值6.0,适宜的酶质量浓度分别为0.20g/dL和0.40~0.60g/dL,温度分别为55.C和65℃,适宜时间都为50-60min;两种酶组合浸提枸杞的较适pH值、温度、时间和酶组合分别为pH值5.5~6.0,55-60℃.50min和0.20g/dL纤维素酶+0.40g/dL木瓜蛋白酶。
