Abstract:用离子色谱法建立一种简单、快速、准确、灵敏的半乳糖醛酸和葡萄糖醛酸的测定方法。多糖水解液经过简单的中和、稀释、过滤后直接进样分析,半乳糖醛酸和葡萄糖醛酸具有较高的分离度(R=4.2);两者在20~200mg/L质量浓度范围内都具有良好的线性,其相关系数均为r=0.9990;重复性和回收率都满足了仪器分析要求;两种化合物的检测限分别为100ng和50ng.

Abstract:采用电击转化方法将双元载体质粒pCAM3300导入根癌农杆菌LBA4404,研究了不同实验条件对转化率的影响.结果表明:当电场强度为11 kV/cm时,转化率最高,每微克DNA得到9.8×103CFU;在细胞OD600为1.0时进行电转化,转化率最高,每微克DNA得到9.54×103CFU;在一定的细胞浓度范围内,电击转化率与细菌浓度密切相关,转化率随着细菌浓度的上升而上升;质粒大小与电击转化率之间没有明确的关系.将含pCAMBIA3300的根癌农杆菌与粗糙脉胞菌共培养转化,在含有膦化麦黄桐(PPT)(405 mg/L)的平板上筛选到转化子.PCR扩增转化子的染色体,初步证明Bar基因整合进粗糙脉胞菌的染色体中.

Abstract:在实验室挤压机上,通过菜籽仁和玉米粉混合挤压,筛选了脱毒添加剂种类,考察了添加剂质量分数、挤压温度、螺杆转速及原料水分对菜籽仁硫甙脱除率的影响.结果表明:菜籽仁和玉米粉混合可实现稳定挤压脱毒;与其它添加剂比较,添加剂(A+B)对硫甙降解脱除效果更显著,硫甙脱除率可高于98%。最合理的混合挤压加工条件为:(A+B)添加量为0.263%+0.179%,挤压温度130℃,螺杆转速95r/min,原料水分质量分数为12.0%。

Abstract:采用稀释平板分离法从土壤中分离到一株产中性蛋白酶芽孢杆菌BX-4.通过单因素碳、氮源以及正交试验确定了最佳产酶培养基配方,同时还研究了起始pH值、金属离子、接种量对产酶的影响以及温度、pH值、金属离子以及表面活性剂对酶稳定性的影响;以最佳培养基为发酵培养基,于37℃、160r/min振荡培养48h,酶活达2519.35U/mL;该酶最适作用温度为55℃,最适作用PH值为7.5,在pH7.5缓冲体系中,55℃反应60min后仍有50%的酶活力,Ca^2+和表面活性剂对酶有一定的激活作用,但激活作用不明显。

Abstract:通过对谷氨酸棒杆菌突变株AATV341进行生物素和VB1添加量的实验,探讨了生物素和VB1添加量对该菌株菌体质量及缬氨酸产量的影响;同时根据对AATV341进行的胞内外某些特定氨基酸和有机酸检测,研究了这两种维生素对碳架代谢流的分布,尤其是对丙酮酸节点处的代谢流量分布的影响。结果表明,生物素添加量在40μg/L时,菌体质量和缬氨酸在胞外的累积明显提高,谷氨酸在胞外累积降低;VB1的适量添加,可提高缬氨酸在胞外的累积,降低发酵液中乙酸和草酰乙酸的含量。

Abstract:采用护色剂ZnCl2和糖(蔗糖、麦芽糖)对冷冻猕猴桃块进行处理,然后分析其在冷冻贮藏过程中的性质变化,实验结果表明:猕猴桃结合糖处理后,其颜色的保持、营养成分的保留以及其玻璃化转变温度与只用护色剂ZnCl2处理的猕猴桃相比,得到了一定的改善,色差中代表绿色强弱的-a值的绝对值提高了近一倍,营养成分维生素C的质量分数也由0.8683%提高到1.1966%,玻璃化转变温度也由-36.818℃提高到-31.345℃.
