Abstract:以少孢根霉RT-3孢子接种在含高浓度的大豆异黄酮提取物制成的发酵基质中,发酵36h后大豆异黄酮被部分水解成相应的苷元.用从丹贝中分离的β-葡萄糖苷酶与标准品染料木素糖苷和大豆苷元的糖苷作用10min,染料木素糖苷和大豆苷元的糖苷被水解成相应的苷元.结果表明:丹贝异黄酮生物活性增强是发酵剂RT-3孢子分泌的β-葡萄糖苷酶将异黄酮由糖苷水解成苷元,而苷元比糖苷有更强的活性.

Abstract:通过采用真空预处理后进行气调包装技术,对生鲜牛肉冷藏过程中一系列品质的变化进行了研究,找到了延长牛肉货架期的较佳条件.主要考察了牛肉在不同的预处理终温、预处理压力及综合优化条件下保鲜效果的优劣.结果表明,选择预处理终温为4℃,预处理压力为1200Pa,联合气调包装充入体积分数40%的O2,25%的CO2气体时,保鲜效果较佳,可使牛肉货架期延长至14d以上.

Abstract:将来源于嗜热脂肪芽孢杆菌的启动子连同β-半乳糖苷酶bgaB基因经PCR扩增后,连接在T载体上,再取代枯草杆菌载体pZ01-bgaB的启动子,将其在枯草杆菌宿主WB600中表达.经摇瓶发酵20h,得到乳糖酶活力6.37U/mL,比活力3.814U/mg,SDS-PAGE电泳显示有明显重组蛋白质条带.证明了嗜热脂肪芽孢杆菌来源的启动子在枯草杆菌中是完全适用的.

Abstract:为了定向筛选α-糖苷酶抑制剂,建立了抑制α-淀粉酶、蔗糖酶和麦芽糖酶活试验为基础的筛选模型.采用此模型筛选从土壤和植物中分离的放线菌,其中菌株E-3519的发酵液对α-淀粉酶和蔗糖酶有强烈的抑制作用.菌株E-3519无气丝,基丝生长好,偶见孢囊,孢囊孢子游动迟缓;能水解淀粉,硝酸盐还原实验阳性,产生硫化氢,能使牛奶冻化;能利用大多数碳源;对枯草杆菌、大肠杆菌、金色葡萄球菌等无拮抗作用;可产生α-糖苷酶抑制剂阿卡波糖.E-3519主要生化特征及形态学特征与ATCC14539相似,命名为Actinoplanes utahensis var.。

Abstract:研究了鹰嘴豆分离蛋白质质量、功能性质、物理化学性质及结构特征.氨基酸分析表明,鹰嘴豆蛋白含有所有必需氨基酸,与FAO/WHO模式相比,色氨酸是其第一限制性氨基酸,蛋白质的化学评分为71分.表面荧光探针ANS法测得的表面疏水性为94.3.DSC热分析显示该蛋白质含有两个热变性温度,分别为83.7℃和95.7℃.圆二色谱(CD)分析了鹰嘴豆分离蛋白的二级结构:α-螺旋36.2%,β-折叠28.4%,转角10.3%,无规卷曲25.2%.经凝胶过滤色谱Sephacryl S-200分离纯化得到7个不同相对分子质量蛋白质组分,其中两个主要组分的相对分子质量为170000及110000.

Abstract:对重组大肠杆菌产β-胡萝卜素发酵条件进行了研究.在M9培养基中添加Span-200.7g/L有利于细胞生长和β-胡萝卜素表达,初始pH值6.0、接种体积分数5%、发酵温度28℃、抗生素氨苄青霉素质量浓度80μg/mL和氯霉素质量浓度40μg/mL为最佳发酵条件,在7L发酵罐进行发酵动力学试验表明,重组菌最适发酵周期为22h,细胞干重最高可达1.55g/L,β-胡萝卜素表达量可达0.75μg/mL。

Abstract:不同厚度薄膜包装对水蜜桃品质变化的影响试验表明,气调包装显著保持果实可滴定酸水平,抑制总糖含量的降低.贮藏温度是影响水蜜桃MAP贮藏效果的重要因素之一.在(3±1)℃经40d贮藏,0.015mmLDPE的MAP失重率显著降低,小于3%,而对照为9.77%.低渗透薄膜(0.03mmLDPE和O.05mmLDPE)MAP果实的生理失调严重,果肉L*值增加.使用厚度为0.015mmLDPE的包装膜在(3±1)℃经40d贮藏,可保持包装内体积分数:O2为3%,CO2为5%.

Abstract:为了测定绿色木霉固态发酵纤维素酶的生物量,采用紫外分光检测固态发酵过程中麦角固醇的量.确定了其最佳提取工艺:在75℃恒温水浴中反应0.5h,用乙醚萃取,乙醇定容,紫外282nm检测.在此基础上对绿色木霉固态发酵的培养基进行了初步的优化,并得到了最佳的培养基组合:碳源为蔗糖,无机氮源为(NH4)2SO4,水料质量比为2.2,pH为5.5.用该培养基,CMC酶活和滤纸酶活分别比优化前提高了21.0%和44.1%.
