Abstract:为构建生物转化法生产茶氨酸的基因工程菌,作者分析了影响大肠杆菌高效表达γ-谷氨酰基转肽酶的几个主要因素.将大肠杆菌JM109的ggt基因接入两种不同的质粒中后,转化大肠杆菌JM109,并在一定的条件下进行表达.通过对比E.coli JM109和转化子的γ-谷氨酰基转肽酶表达活性的不同,探讨了ggt基因的拷贝数、启动子的强弱等几个因素对γ-谷氨酰基转肽酶高效表达的影响.实验表明,将含有自身信号肽和启动子的E.coli JM109的ggt基因接入高拷贝的质粒pUC18中,仅提高ggt基因的拷贝数不能增加γ-谷氨酰基转肽酶的表达量.将含有自身信号肽但不含有自身启动子的E.coli JM109的ggt基因接入具有tac启动子的表达载体pEtac中,发现在强启动子tac的控制下,γ-谷氨酰基转肽酶的表达量提高至出发菌株的2.3倍.将构建的工程菌用于生物转化法生成茶氨酸,底物转化率相应地提高至出发菌株的1.7倍.

Abstract:在含50mL/L的甲醇、5g/L的甘氨酸的基本培养基平板上,从土壤中分离出130多株细菌,测定了它们利用甘氨酸生产L-丝氨酸的能力,获得了一株产L-丝氨酸较好的菌株A3,L-丝氨酸产量最高为1.4g/L,分类学鉴定为微球菌科(Micrococcaceae),对菌株A3利用甘氨酸发酵生产L-丝氨酸的发酵条件进行了初步研究,包括甘氨酸和甲醇添加量,不同碳源、氮源,初始pH值以及发酵时间对产酸的影响.结果表明,菌株A3在不添加甲醇、只以甘氨酸为惟一碳源的发酵培养基中也能产L-丝氨酸,培养基中较高质量浓度的甘氨酸存在对L-丝氨酸生产是必需的.添加葡萄糖、甘油等外源碳源对菌体生长有利但不利于L-丝氨酸的生产。

Abstract:研究了超声场下胰蛋白酶水解酪蛋白的酶解反应条件与动力学.在超声波频率 20 kHz、作用与间歇时间比为1∶1时,测定了不同功率下胰蛋白酶活性及该酶的最适温度、最适 pH和米氏常数Km 的影响.结果表明,180,225 W功率下,超声10 min的酪蛋白水解率显著高于常规对照组(P<0.05); 225 W超声功率下,在5 ℃,10 ℃,20 ℃下水解率分别比对照组提高 4.4 倍、2.47倍和2.33倍(P<0.01);超声场下胰蛋白酶 Km 值减小,Vm 增大.将胰蛋白酶、底物分别进行超声处理,紫外和荧光光谱分析观察到,超声处理后,酶溶液荧光发射光波长从334 nm移至332 nm,荧光强度提高,表明一定的超声波可能使酶及底物分子构象发生变化,提高胰蛋白酶水解活性.

Abstract:冷冻甘薯片在贮藏过程中发生的褐变与多酚氧化酶(PPO)的活性呈正相关.实验研究了甘薯片中多酚氧化酶的性质,并采用各种护色剂抑制甘薯片在冷冻加工过程中产生的褐变.结果表明,甘薯中多酚氧化酶在70℃下很不稳定,随着恒温时间的延长,活性迅速降低;甘薯中多酚氧化酶的最适pH值分别为5.0和7.5;甘薯片的最佳护色方法为热处理结合护色剂,L-半胱氨酸是较理想的护色剂,它不仅能够很好地抑制甘薯片在冷冻过程中发生的褐变,而且对甘薯的营养成分如VC具有保护作用。

Abstract:本试验研究了在经膨化处理的陈化早籼糙米日粮中添加外源α-淀粉酶和糖化酶对断奶仔猪小肠黏膜二糖酶、血液生化指标和生产性能的影响.试验为日粮不同加工处理(膨化、不膨化)和加外源酶(添加、不添加)两因子设计,共4个处理.结果表明,试验猪的生产性能总体趋势是非膨化组好于膨化组,非膨化加酶组的日增重比膨化加酶组在第14天和第28天分别高出23.66%和16.83%,料肉质量比降低了16.88%和7.36%;膨化和添加外源酶显著提高了断奶仔猪试验第14天十二指肠中麦芽糖酶的活性,然而膨化显著降低了断奶仔猪试验第28天空肠中乳糖酶、麦芽糖酶、异麦芽糖酶和蔗糖酶的活性,添加外源酶显著提高了断奶仔猪试验第28天空肠中乳糖酶和麦芽糖酶的活性.试验结果表明,对断奶仔猪高糊化度膨化陈化糙米是没有必要的.
