Abstract:采用超临界CO2萃取技术对番茄籽油进行萃取,经过单因素和优化实验,对不同萃取时间、压力和温度下油的萃取率、脂肪酸组成和品质进行了比较.其中萃取时间和萃取压力对番茄籽油的萃取率影响显著(α=0.05),萃取温度影响较显著(α=0.1).最佳萃取条件为萃取时间2 h、萃取温度50℃、萃取压力30 MPa,萃取率达96.34%.通过气相色谱分析,由超临界CO2萃取得到番茄籽油的主要脂肪酸质量分数分别为棕榈酸(C160)约13%,硬脂酸(C180)约5%,油酸(C181)约22%,亚油酸(C182)约56%,花生酸(C200)约2.8%.通过测定番茄籽油过氧化值、酸价、碘价和皂化价,显示番茄籽油有较好的品质,不同的萃取条件对番茄籽油品质无显著影响.

Abstract:FML02-8分别以葡萄糖、果糖、乳糖、蔗糖、麦芽糖、乳清粉为碳源合成不同量、不同结构的胞外多糖,经红外光谱分析,波形、特征吸收波段、波峰强度均有差别;用HPLC法测得的相对分子质量分别为41092,54 029,45 468,46 996,46 257,45 468;用气相色谱法分析得到的单糖组成及摩尔比均不同.结果表明,碳源不仅影响FML02-8合成EPS的产量,而且影响EPS的结构,包括相对分子质量、单糖组成及摩尔比等.

Abstract:对α-葡萄糖醛酸酶基因(AguA)的稀有密码子进行了优化,经过稀有密码子优化的基因在pTrc99A载体中由于二级结构稳定性的增加使得目的基因的表达水平比原始基因还要低.在权衡几个载体后使用本实验室构建的热激载体pAT,可以降低mRNA二级结构自由能(△g),得到了α-葡萄糖醛酸酶的高表达,结果充分说明了mRNA二级结构的自由能对外源基因的表达的影响.

Abstract:通过对溶壁微球菌培养基优化,确定最佳发酵培养基,并对发酵工艺条件进行了初步的优化.最适碳源是麦芽糖,最适氮源是蛋白胨和酵母膏,最近无机盐是MnSO4;该菌株产酶的最佳培养基配方为:60 g麦芽糖,13 g蛋白胨,8 g酵母膏,1 g MnSO4,5 g NaCl,2 g NaH2 PO4·2H2O,2 g K2HPO4·3H2O,0.5 g MgSO4·7H2O,0.5 g CaCl2,1 000 mL水.从该菌株发酵过程曲线发现,该菌株大量产酶期在12-14 h.摇瓶发酵最适初始pH值为7.0,250 mL三角瓶装液量为25mL,接种体积分数为5%,培养温度为37℃.

Abstract:通过对过氧化物酶的残留酶活、色差、剪切强度和剪切功的分析,研究了热烫时间和贮藏温度对蒲菜的过氧化物酶、色泽和质构的影响.结果表明,在100℃的热烫温度下随着热烫时间的延长,过氧化物酶活降低,而贮藏2 d后酶活的再生量先增大后降低,在热处理时间为1.0-1.5min时达到最大值;热烫时间为4 min时,可基本破坏蒲菜中的过氧化物酶,且蒲菜的质构和色泽较好.

Abstract:对美味牛肝菌(Boletus edulis)深层发酵条件及菌丝体主要营养成分进行了分析研究,结果表明,最适培养基配方为:葡萄糖30.0 g,玉米粉20.0 g,酵母膏7.0 g,KH2PO4 2.0 g,MgSO4·7H2O 1.0 g,VB1 0.01 mg,CaCO3 2.0 g;最优发酵条件的初始pH值5.2-5.8,振荡速度180 r/min,培养温度27℃,100 mL三角瓶装液量60 mL.并对美味牛肝菌深层培养菌丝体中蛋白质成分、灰分、脂肪、粗纤维、矿物质及多糖等进行了测定和分析.

Abstract:探讨了胚芽乳杆菌在鳕鱼下脚料水解液(4 g/L葡萄糖)中的生长及产酸情况,结果表明,在32℃的恒温室,经过24 h的静止培养,水解液的pH值为4.3,菌落数(CFU)为4.0×109,低于相同条件下在MRS培养的pH值3.8,菌落数(CFU)5.5×109;但在水解液中培养至72 h时,水解液的pH值可降至4.1.在水解液中静止培养的前20 h是胚芽乳杆菌生长的旺盛期,连续培养应从静止培养15 h、pH值4.5左右开始.在连续培养的105 h内,新鲜培养基的流加速度为70 mL/d,pH值几乎能恒定在4.5左右,葡萄糖和乳酸含量也基本保持恒定.
