Abstract:脂肪酸和脂肪酸酯是呈“酸性”的,它是使发酵法生产丙三醇的NaOH滴定值超标的一类杂质.为分析其产生来源及进一步提出控制策略,确定了显“酸性”的关键脂肪酸酯.GC-MS分析发现,发酵法生产工业级丙三醇中脂肪酸及脂肪酸酯为乙酸、丙酸、丁酸甘油酯及甘油单乙酸酯等,其中甘油单乙酸酯含量最高.HPLC法测定了不同厂家发酵生产的工业级丙三醇中甘油单乙酸酯的含量,一般在0.5~3.0g/100g丙三醇.它对工业级丙三醇的脂肪酸滴定值影响最大,是发酵法生产工业级丙三醇显“酸性”的关键物质.

Abstract:选择122树脂用于丙酮酸发酵液的脱色,在发酵液丙酮酸质量浓度为60g/L、pH3.0、脱色空间流速为1.8mL/(mL*h)的条件下可取得较好的脱色效果,丙酮酸损失率低于6%.从8种碱性阴离子交换树脂中筛选出330弱碱性阴离子树脂(氯型),用于从发酵液中提取丙酮酸.考察了不同操作条件对固定床离子交换效果的影响,上柱料液pH的变化对流出曲线的形状影响不大.当上柱料液的pH和丙酮酸质量浓度分别控制在4.0和40g/L时,可以获得较高的有效体积交换容量(分别为0.534mol/L树脂和0.530mol/L树脂).实验范围内体积流量对交换过程的影响不大,在较低的离子交换空间(0.44mL/(mL*h))下有效体积交换容量较高(0.604mol/L树脂).考察了固定床洗脱工艺条件对洗脱效果的影响.当洗脱剂盐酸的浓度为2mol/L、洗脱空间流速控制在0.44mL/(mL*h)左右时洗脱效果较好.采用较佳的工艺条件进行固定床单柱操作,实验得到离子交换单元的丙酮酸收率为97.0%.

Abstract:主要研究了机械破壁、pH值以及麦芽根核酸酶的作用对啤酒酵母自溶过程的影响.均质破壁能显著提高自溶速率及酵母抽提物中蛋白质和核苷酸的含量,麦芽根核酸酶的加入有效地提高了酵母抽提物中5′-核苷酸含量,而提高自溶时的pH值不能显著地提高酵母抽提物中5′-核苷酸的含量.因此,有效地提高酵母抽提物中5′-核苷酸含量的方法是啤酒废酵母经0.5%NaHCO3脱苦后,加2倍体积无菌水和5%NaCl,再加质量分数为1%的核酸酶,搅拌均匀,在40MPa下均质3次,然后升温至55℃,自溶30h,得到的酵母抽提物中5′-GMP含量高达6.37mg/g.

Abstract:对在不同破碎温度下所得番茄浆的动态流变性质以及果胶物质相对分子质量的分布和大小进行了研究.结果表明随着破碎温度的增加,番茄浆中果胶物质的平均相对分子质量增加.但当破碎温度为90℃时,果胶物质的平均相对分子质量却有所下降.在80℃和85℃破碎时,番茄浆具有相当大的贮能模量,而在75℃和90℃破碎时,贮能模量值较小.贮能模量值对振荡频率具有一定的依赖性,故番茄浆表现出弱凝胶特性.贮能模量G′和损耗模量G″交汇点的位置随着破碎温度的升高向低频区移动.这说明破碎温度越高,大分子物质被保留得愈多,体系呈较强的弹性;破碎温度越低,体系中大分子物质被保留得越少,体系呈较强的粘性.
