Abstract:1.75%NaCl溶液可有效抽提林木毛霉菌丝体表面的蛋白酶,依此确立了腐乳毛坯中蛋白酶的检测法。该酶系在40%以下2h酶活稳定不变,60℃lh能保持58%活力;其最适作用温度和pH分别为55~60℃和6~7pH;pH稳定范围为4~7;NaCl和CH_3CH_2OH对酶系有明显的保护作用。讨论了蛋白酶检测及其性质对腐乳工艺改进的指导意义。

Abstract:从毛霉MucorZ3l8出发,选育出M.Z328变异株。研究了M.Z328变异株的前酵条件:T=30±1℃,相对湿度95%.采用新工艺进行生产实验,前酵时间仅需22h.首次发现毛霉菌丝能渗透到豆腐坯的最深层。用正交试验确定后酵条件。优化结果表明,采用低盐控温后酵新工艺,能将后酵时间缩短到10~12d,即可生产出氨基酸态氮高达1.75g/100g干腐乳以上的白腐乳。其质构、颜色等均能达到质量要求。研究了腐乳发酵过程的流变特性,微细结构及DSC.图谱的变化情况。
