    Abstract:

    The purification,components analysis and hypolipidemic activity of a polysaccharide from Camellia oleifera Abel. seeds(CPSI) were studied. The crude polysaccharides were extracted from the seeds of Camellia oleifera Abel. using deionized water. The Camellia oleifera seeds polysaccharides(CPS) were further separated and purified by anion exchange chromatograph on DE-52,Sephadex G-100 column and affinity chromatography. The monosaccharide composition in the Camellia oleifera seed polysaccharide Ⅰ was determined by capillary electrophoresis.The potential mechanisms underlying lipid-lowering affect of CPSI were examined,specifically alterations in levels of the total cholesterol(T-CHO),High-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),3-hydroxy-3-methyl glutaryl coenzyme A reductase(HMG-CoA) and coprosterol,using a mice model of hypercholesterolemia. CPSⅠwas conjugated polysaccharide which has many phosphate groups. The polysaccharide contained xylose and galactose,and the mole ratio of galactose and xylose was 4.6∶5.57 in the hydrolytic solution of CPSⅠ.Serum T-CHO,LDL-C levels in mice treated with CPSI declined significantly when compared to hyperlipid control group,and coprosterol of mice in the groups treated with CPSI was higher than that in hyperlipid control group.Serum HDL-cholesterol and HMG-CoA activity,however,was unchanged.These results indicate that CPSI is useful for the prevention of hyperlipidemi in mice and it may therefore play a valuable role in reducing cardiovascular risk.
