Identification of araE Gene Associated with Response to Different Phenylpropanoids by Gene Silencing in Escherichia coli BL21

    Abstract:

    AraE is an important unidirectional transport channel protein that could transport some compounds outside the cell. E. coli is a common host-bacterial to produce phenylpropanoids,and the growth was inihibited by phenylpropanoids. In order to investigate the transport ability of AraE on phenylpropanoids in E. coli,the gene overexpression and silencing routes was used in this study to regulate the expression of AraE. The level of overexpression and silencing was confirmed by qPCR and SDS-PAGE. The transport ability of phenylpropanoids was measured by detection of the recombinant bacterial growth kinetics parameter (μmax) in different typical phenylpropanoids (rutin,naringenin and resveratrol). The result showed that AraE could only transport resveratrol in E. coli.
