    Abstract:

    In order to obtain the optimal medium composition for Pullulan production,statistical method was used to investigate the medium components for pullulan concentration by Aureobasidium pullulans in this study First,the Plackett-Burman design was used to evaluate the effects of six variables including glucose,yeast extract,KH2PO4,MgSO4 7H2O,(NH4)2SO4 and NaCl.Subsequently,a three-level three-factor Box-Benhnken design was employed to determine the maximum Pullulan production at optimum concentration of glucose,yeast extract and NaCl.The optimal values of the three parameters determined by RSM were glucose 5.675%,yeast extract 0.405% and NaCl 0.0815%.The Pullulan production was 32.1g/L,and the actual experimental result was 32.16g/L.They were in well agreement and it is 5.0 folds of the Pullulan production before optimization.
